2 Collision cell: Fragmentation efficiency > 90 % Duty cycle: Variable between 1 and 50 % (user selectable trapping time/scan time ratio) Transmission: approximately 100 % MS/MS Sensitivity, ESI: A loop injection of 50 pg Erythromycin shows a signal to noise of better than 10/1 for the four most abundant daughter ions (parent ion [M+H] = ; scan range 250 to 750 Da). Mass Spectrometer (MS-1): a. New computer calculated wide angle ion optics of reverse Nier-Johnson geometry. The optics is optimized for an acceleration voltage of 5 kv. All ionization techniques are operated with full acceleration voltage. b. Constant mass calibration based on magnetic field measurement. Calibration is independent of mass range, scan speed, scan mode (including linked scan MS/MS) and ionization technique. c. Efficient temperature regulated ion source assembly for use with all ionization techniques, including one EI ionization volume with filament. d. Quadratic ion source manifold with multiple inlet ports for high flexibility. e. Radially laminated, computer designed H-type magnet with mass independent focal length. The radius of deflection is 350 mm, the angle of deflection is 65. f. Electrostatic analyzer (ESA) of advanced design. All focusing and detection elements are assembled on an optical bench and are housed in a single vacuum chamber. g. Off axis secondary electron multiplier detection system with postacceleration/conversion dynode, variable to +/- 20 kv (Finnigan MAT patent). h. The entire analyzer is assembled on an integrated shock mounted platform for isolation from floor vibration. 2

3 i. Directly coupled high speed differential pumping system with three turbo molecular pumps, push-button control and automated protection system. j. Vertical electronics cabinet with optimal ventilation and easy access for maintenance, containing all analog supply and drive electronics. k. Digital electronic card cage with Pentium III processor and 64 MByte memory for instrument control (ion source, inlet system, analyzer and scan control, display control and system communications). l. Optocoupler isolation between digital electronics and mass spectrometer high voltage and power supplies. m. Universal input/output board for readout and control of external devices, (e.g. UV detector) with 1 analog IN, 1 digital IN and 1 digital OUT, under full ICL control. Mass Spectrometer (MS-2) a. High efficiency deceleration- and dual octapole ion transport optics. b. IonTrap with axial modulation for optimized accuracy and reproducibility. c. MS/MS n capability (MS>MS>MS> experiments with highest sensitivity). d. Off axis secondary electron multiplier detection system with postacceleration/conversion dynode, variable to +/- 15 kv (Finnigan MAT patent). e. Turbomolecular pump with mechanical forepump. f. Full data system control. g. The intermediate detector system for normal BE operation in MS 1 remains unaffected by the IonTrap analyzer. API Interface: k. Patented heated metal capillary for efficient ion transfer and complete sample desolvation. l. Patented lens between metal capillary and skimmer for improved sensitivity. m. RF-quadrupole transfer optics for improved ion focussing and sensitivity in CID- MS. 3

Electrospray: r. Patented needle design with built-in liquid sheath for increased performance with 100 % aqueous mobile phase and strong buffers. Accommodates liquid flows of 1 µl/ min to 1 ml/min without splitting. Atmospheric Pressure Chemical Ionization (APCI) Unique heater/sensor design ensures efficient heat transfer to liquid effluent. Accommodates liquid flow up to 2 ml/min without splitting (requires ). Micro Electrospray Ionization (Micro-ESI) Unique sprayer for pneumatically infusion at nano liter flow rates with manual xyz-adjuster. Accommodates liquid flow from 20 nl to 1µl/min. (requires ). Continuously (flow) adjustable and heatable reference compound and CI reagent gas inlet system. Allows syringe introduction of liquid or gaseous samples d. Direct insertion probe, computer controlled, liquid cooled, indirectly heated with a maximum temperature range of 350 C. Heating rates 20 C to 200 C in 30 seconds/200 C to 300 C in 30 seconds e. Trace 2000 GC with GC Coupling Parts High performance capillary gas chromatograph (GC) Trace 2000 with electronic pressure control (EPC) and multiple method storage. GC can be operated independently or via fully integrated data system control. High performance split/splitless capillary injector. Direct coupling GC/MS interface with uniform temperature distribution and precise temperature control up to 400 C. GC PAL Autosampler for Trace 2000 GC Reliable stepper motors positions the syringe carriage above each vial for sampling. Capable of handling low volume samples (triplicate 1 µl injection from as little as 10 µl of sample microvials). The autosampler is under full ICL control.
