The eye can scarcely see them in bright field and dark field. However, very small differences exist between the refractive indices of the cells and the surrounding aqueous solutions and within the cells between the cytoplasm and the cell nucleus. The higher the refractive index of a medium, the smaller the speed or velocity of light in the medium. It translates the tiny differences into differences in intensity.
