The software version in ZEN Help About must be 14.0.x.201 Please contact your local service engineer prior to updating a customized system (SCS or INDIMO system). Please contact your local service engineer prior to updating ELYRA systems with newly installed Andor SDK require a database adjustment. The Hotfix may not be installed on systems running on a different ZEN version than specified above. Installation instructions: Run the installation file using a Windows user account with administrator privileges. The ZEN software must not be running during the installation. New Features introduced by Feature Pack 1, which was included in a previous Hotfix: Experiment Designer Added the option to configure an Interval block. This block will delay the acquisition of the subsequent block for a specified time, while taking into account the time required for the previous block. (#159068) Experiment Designer The experiment blocks will show an indication of the time required for their respective acquisition. (#159068) Issues addressed by this Hotfix: General - Solved an issue, where Auto Z Brightness Correction for Z-Stack Experiments using NLO Laser shows an increase of laser intensity. (#160415) General - The "Test Autofocus" button will no longer affect the scan speed of Z-Stack experiments. (#164410) General - For multitrack experiments with selected mode "Frame Fast", when changing Optovar positions, now changes for all tracks automatically. (#164520) 1

(#169757) General - Solved an issue for Z-Stack Experiments with Auto Z Brightness Correction, where the correction was done within individual images. (#171031) Experiment Designer - "Bleach in different Z" check box now works correctly in multi-block experiments. (#169270) Airyscan - Solved an issue where ZEN has stopped working in a bleaching experiment. (#162593) Airyscan - Fixed stability issues when changing the sampling regime or Frame Size while the Airyscan view is active. (#174458) Airyscan Fast - Solved an issue where some images were under certain circumstances not saved completely. (#162367) ELYRA - PALM datasets processed before ZEN 2.3 SP1 HF4 will be opened correctly. (#151518) ELYRA - Model-based drift correction now recognizes markers in the image in adaptive mode for automatic range detection. (#157826) Piezo Z-Drives - Improved precision of the step size calculation. (#154974) Piezo WSB Actively stopping Experiments by clicking the stop button will no longer cause randomly occurring Z-offsets. (#162640) Piezo WSB Corrected a significant offset in the Z-Stack range used by the piezo compared to the internal focus drive. (#162971) Piezo WSB Fixed an issue where Z-Stacks of one tile are not in the correct order. (#164304) Piezo WSB Corrected a significant offset in the Z-Stack range used by the piezo compared to the internal focus drive when using Frame or Frame Fast multi-tracks. Issues addressed by previous Hotfixes and included in this Hotfix: General - External Airyscan processing of Time Series will now be initiated without a delay. (#159782) General - The interval of time series is correctly maintained when tracks are added or removed. (#157567) General - On systems with switchable Optovar, the Optovar will now be correctly kept stable during multitrack acquisition with Frame Fast track switching. (#154772) Airyscan - Solved an issue with Airyscan processing of images, which contain T-PMT channels. (#158222) Airyscan - Improved stability of Airyscan batch processing with large number of files. (#160213) Airyscan Fast - Fixed an issue, which could cause equally spaced black lines in the image under certain sampling conditions. (#154302) Airyscan Fast - Fixed the problem that Bleaching ROIs could eventually be disappearing when the experiment was cancelled by the user. (#154896) Experiment Designer - Duplicating Experiment Blocks does not alter the activation status of inactive tracks any longer. (#154545) Ortho Viewer - The controls to move graphical elements based on pixels have been removed. (#153207) 2

When multitrack acquisition of time series with frame-wise averaging is performed, the acquisition will now maintain a constantly high speed. (#139868) Airyscan - When Airyscan Fast is used to acquire Tile scan images after an overview image has been generated with a different objective or certain acquisition settings, the image is now displayed correctly. (#151057) Airyscan - For Airyscan Fast systems on Axio Examiner or an Axio Observer (rear-port) stands, the option "exchange XY" was enabled by default with every start of ZEN. As a consequence of this option, the image orientation in the ocular matches the image display in the software. However, the scan area representation of non-symmetrical scan fields is flipped on Airyscan Fast acquisition on these systems, including the scan area offsets and non-symmetrical tile scans. By deactivating the option "exchange XY" the issues in the scan area representation and in the tiles acquisition can be avoided. This Hotfix changes the behavior of the "exchange XY" checkbox, so that the setting will be maintained when ZEN is restarted. (#150894) Airyscan - multitrack Live acquisition will maintain its speed. (#156321) Airyscan - Improved robustness in multitrack experiments after switching an Airyscan track to LSM and then switching to the Airyscan viewer. ELYRA - Fixed an issue which prevented 3D PALM datasets to be rendered for the entire stack height. (#135385) ELYRA - Improved the performance of SIM acquisition, which was slower in this Service Pack version compared to previous ZEN releases. (#149379) ELYRA - On systems with MiCo interface HS (not on systems with real-time PC) long exposure times of >400ms can now be used for multitrack SIM acquisitions. (#148790) ELYRA - Fixed an issue where the time series acquisition occasionally stopped with PALM sequential mode and trigger mode internal. (#160601) ELYRA - In SIM billing (at 405 nm, sometimes at 488 nm) levels no longer appear black. ELYRA - Multitrack laser WF experiments will no longer show a delay in dependence on the laser power. (#146030) ELYRA - The metadata regarding the camera zoom factor were misleading and therefore removed from the info view. (#132894) Analysis - The apparent molecular brightness B is now scaled according to the unit photons/pixel in compliance with literature instead of khz. For immobile structures B will now equal to 1. (#141626) Cameras - When multitrack wide-field images are reused, the exposure times are now correctly set and the will also not be restricted any more. (#132679) LSM More than one dye spectrum can be displayed in the Imaging Setup for Lambda mode acquisition.
