1 OUTLINE Distinguishing populations of living and, using a non-invasive opto-acoustic analysis of wing beat patterns. Counting Mosquitoes Distinguishing gambiae s.l. Outlook Andreas Rose 1, Sergej Sperling 1, Ulla Gordon 1, Michael Weber 1, Matthias Geismar 2, Ilyas Potamitis 3,, Pancraç Villalonga 4, João Encarnação 4, Martin Geier 1 1 Biogents AG, Regensburg, Germany 2 University of Regensburg, Regensburg, Germany 3 Technological Educational Institute of Crete, Rethymno, Greece 4 Irideon S.L., Alaior (Baleares), Spain * andreas.rose@biogents.com 1 2 Counting Mosquitoes Counting Mosquitoes BG-Counter BG-Sentinel 2 CO
2 REMOSIS project goal Make it species-specific (Since February 2016) Distinguishing and 7 8 Wider Sensor Field: Ilyas Sensor Rearing & An. : Same densities in containers for larvae & pupae Same densities in bugdorms Same age 9 10 Result Exp. 1: Populations Experiment 1: Measuring Populations 2 bug dorms, one with 30 females of either species Sensor into bugdorm 1, breathing into bugdorm every 2 minutes After 15 minutes, change SD cards, Median 528 Number of recordings : 3103 Median 573 Number of recordings: 3093 N=3103 N=3093 move sensor into bug dorm 2 Repeat, until each bugdorm / species has been measured 60 minutes Difference between the medians wing beat frequency: An. 45 Hz lower
3 Experiment 2: Measuring Individuals Each mosquito from each bugdorm from experiment 1 was placed individually into a small flight chamber with the sensor. Breathing into chamber to stimulate flight Record 5 flight tracks File mosquito in marked collection Measure wing length 13 N=256 N= gambia s.s gambia s.s Difference between the medians wing lengths: An. 139 µm longer Difference between the medianswing beat frequency: An Hz lower (compared to 45 Hz from experiment 1) , same wing lengths n=83 n= Wing length gambia s.s
4 True Species, same wing lengths gambia s.s Difference between the medianswing lengths: An. 4 µm shorter (compared to 139 µm longer in the population measured in experiment 2) Difference between the medianswing beat frequency: An. still 42,5 Hz lower (compared to 41.4 from the total population measured in experiment 2, and 45 Hz from experiment 1) Laboratory-reared had a by 41.4 to 45 Hz lower wing beat frequency than laboratory-reared Laboratory-reared had149 µm longer wingsthan laboratory-reared The difference in wing beat frequencies can however not be attributed to the differences in wing length: Comparing two populationswith the same wing length still exhibited the difference in wing beat frequencies (42.5 Hz) And now Ae. aegypti Ae. albopictus Cx. pipiens Cx. quinquefasciatus An. An. Total of 280,000 tracks Confusion Matrix Thank you! The development of the BG-Counter was partly supported by the EU's 7th Framework Programme (grant , acronym MCD) The continuation of the development is being supported by the EU's Horizon 2020 programme (grant , acronym REMOSIS).
