1 INSTRUCTIONS IX2-MLWCD MID-LONG WORKING DISTANCE CONDENSER This instruction manual is for the Olympus Mid-Long Working Distance Condenser Model IX2-MLWCD. To ensure the safety, obtain optimum performance and to familiarize yourself fully with the use of this equipment, we recommend that you study this manual thoroughly before operating the system. Retain this instruction manual in an easily accessible place near the work desk for future reference. A X

4 IMPORTANT This product is a condenser with mid-long working distance (WD 45 mm, NA 0.5) designed for use in the OLRC (Olympus Relief Contrast) observation. It is also compatible with other observation methods including the brightfield, phase contrast, DIC (Differential Interference Contrast) (20X or 40X) and simplified polarized light observations. When the condenser is combined with the DIC polarizer unit (IX2-MLWPO), it is possible to switch between the OLRC and DIC observations quickly. With the OLRC observation, the orientations of the shadows even at different magnifications are constant. This makes observation easy even when the magnification is changed, and the maintenance of an equivalent focal distance minimizes defocusing resulting from magnification changes. 1 Getting Ready 1. This manual pertains only to the mid-long WD condenser. Please also read the instruction manual for the IX2 series (IX81/ IX71/IX51) or IX series (IX70/IX50) microscope to understand the comprehensive operating instructions of the microscope system. 2. For the illumination column, only the IX2-ILL100 is applicable. Also be sure to use the dedicated phase contrast and DIC optical elements. 3. The mid-long WD condenser is a precision instrument. Handle it with care and avoid subjecting it to sudden or severe impact. 4. Be careful not to leave dirt or fingerprints on the lens components. 5. The stopper mechanisms provided for the functions indicate the limits of motion. Never attempt to go beyond a stopper by applying excessive force. 6. Do not tighten the DIC slider clamping screw too much. 1

5 IX2-MLWCD 2 Maintenance and Storage 1. To clean the lenses and other glass components, simply blow dirty away using a commercially available blower and wipe gently using a piece of cleaning paper (or clean gauze). If a lens is stained with fingerprints or oil smudges, wipe it gauze slightly moistened with commercially available absolute alcohol. Since the absolute alcohol is highly flammable, it must be handled carefully. Be sure to keep it away from open flames or potential sources of electrical sparks --- for example, electrical equipment that is being switched on or off. Also remember to always use it only in a well-ventilated room. 2. Never attempt to disassemble any part of the equipment, for this may degrade the performance. 3 Caution If the condenser is used in a manner not specified by this manual, the safety of the user may be imperiled. In addition, the equipment may also be damaged. Always use the equipment outlined in this instruction manual. 8 ASSEMBLY }This chapter describes the assembly for OLRC (Olympus Relief Contrast) observation. The assembly methods for other observation methods will be described in their respective sections. 5 Fig. 1 ƒ Fig. ² ³ 1 Attaching the Modulators (Fig. 1) }The three modulators can be attached to the three positions other than the BF (Brightfield) observation light path. 1. Loosen the clamping and remove the cover ². 2. Rotate the turret ³ to engage the position you want to insert the first modulator in the light path. 3. Insert the two centering knobs into the two centering knob insertion holes (marked ) and loosen the internal adjustment screws slightly (by turning them counterclockwise). # Do not loosen the adjustment screws too much, for this causes the adjustment screws to project out of the turret and contacts the turret cover, thereby hindering the rotation. 4. Rotate the turret by 180 to the position of the cover. Now, while pushing the spring ƒ obliquely with the modulator, insert the modulator in the orientation as shown in the figure; this will facilitate the later adjustment operation. 5. Check the hole position number and attach the magnification index to the correct number position on the front of the condenser. (Magnetic absorption) 6. Rotate the turret by 180, access the adjustment screws in the same way as step 3 and tighten them lightly. 7. Attach the two other modulators in the same way as above. }Attach the BF index to the position without modulator. 8. After attaching the modulators, attach the cover ².

9 IX2-MLWCD 2 Attaching the Dedicated Polarizer (Fig. If the dedicated is not attached yet, screw it into the top of the condenser. Fig. 3 3 Attaching the IX2-MLWPO Polarizer Unit (Fig. 4) ³ ² Fig. }If this polarizer unit is not used, the contrast in DIC observation may be degraded. 1. Turn the dedicated polarizer on the top of the condenser counterclockwise to remove. 2. Fit the IX2-MLWPO into the stop of the condenser so that the polarizer rotation/clamping screw ² comes on the front as shown in the figure. (Attach by aligning the positioning pin and groove.) 3. Tighten the clamping screw ³ firmly using the Allen screwdriver. 4. Screw in the dedicated 6

10 4 Attaching the Mid-Long Working Distance Condenser Attach the mid-long WD condenser on the condenser holder by referring to the instruction manual for the IX2 series (IX81/IX71/IX51) or IX series (IX70/IX50) microscope frame. 5 Centering the Mid-Long Working Distance Condenser 1. Disengage the DIC slider and analyzer from the light path, and rotate the turret of the IX2-MLWCD to select the BF light path. The dedicated polarizer can be left attached to the condenser because it does not cause a trouble even when it is engaged in the light path. 2. Center the condenser by referring to the instruction manual for the IX2 series (IX81/IX71/IX51) or IX series (IX70/IX50) microscope frame. 7

11 OBSERVATION METHODS IX2-MLWCD 3-1 Brightfield Observation }Objectives with magnification from 4X to 40X can be used. 1. Disengage the analyzer and DIC slider from the light path, and set the turret for the BF light path (the position without modulator, DIC prism and phase contrast ring slit). }Brighter observation is possible if the dedicated polarizer is removed, but this can also left attached. 2. Engage the 10X objective in the light path and bring the specimen into focus. 3. Increase the objective power step by step and bring the specimen into focus every time; repeat this until the target magnification is reached. 4. Adjust the field iris diaphragm (FS lever) so that it circumscribes the field of view. }If the centration of the field iris diaphragm has not been adjusted, adjust it by referring to the instruction manual for the microscope frame. 5. Adjust the aperture iris diaphragm (AS lever) properly. 8

12 3-2 OLRC (Olympus Relief Contrast) Observation }Use an OLRC-dedicated objective. 1 Adjusting the Modulator (Fig. 5) ² ³ Fig. }In the adjustment stage, it is recommended to use a general dyed specimen in order to facilitate the focus adjustment. If such a specimen is not available, use dirt or scratch on the petri dish for focusing. }Open the field iris diaphragm (FS lever). 1. Rotate the to engage the desired modulator in the light path. 2. Engage the objective with the same magnification as the above modulator in the light path and bring the specimen into focus. 3. After obtaining the focus, remove the specimen, remove an eyepiece and look into the eyepiece sleeve. }When the U-BI90CT binocular observation tube is used, rotate the CT turret to position CT to engage the CT lens in the light path. When the U-BI90 or U-TBI90 binocular observation tube or U-TR30H trinocular observation tube is used, remove an eyepiece and attach the U-CT30 centering telescope. 9

13 IX2-MLWCD B A Dark area Gray area Bring the objective s exit pupil into focus in the following method. U-BI90CT: Turn the focus adjustment ring. U-CT30: Turn the upper adjustment ring. 4. The objective s exit pupil looks as shown in Fig. 6. Overlap A of the modulator with the gray area of the objective using the centering knobs. A: Area where light is transmitted permanently. B: Area where the transmittance varies according to the rotation of the dedicated polarizer. Circumferential-direction movement: Insert a centering knob directly into the modulator s groove through the modulator rotation groove ² and turn the knob. (Fig. 5) XY-direction movement: Insert the two centering knobs into the two insertion holes ³ and turn the knobs. (Fig. 5) 5. Also adjust the modulators with other magnifications. Fig. 6 10

14 2 OLRC Observation Procedure }When the CT turret is in use, set it to position indicating \. When the U- CT30 is in use, replace the eyepiece. 1. Place the specimen on the stage. 2. Engage the desired objective and modulator in the light path and bring the specimen into focus. 3. Adjust the field iris diaphragm (FS lever) so that its image circumscribes the field of view. 4. Rotate the dedicated polarizer for contrast adjustment to obtain the optimum contrast for the specimen. This enables 3D type observation of a thick, non-dyed specimen on a uniform, gray background. 5. As the 20X objective has an aperture iris diaphragm effect, adjust it properly. With the 10X and 40X objectives, simply open the aperture iris diaphragm. 11

15 IX2-MLWCD 3-3 DIC (Differential Interference Contrast) Observation }If a plastic petri dish is used, the DIC effect cannot be obtained due to the polarization characteristic of the plastic material. Be sure to use a glass petri dish. }The DIC observation requires, in addition to the IX2-MLWCD mid-long WD condenser, the dedicated DIC prism, the U-DICTS DIC slider (to be used together with the IX2-AN analyzer) or U-DICT DIC slider (to be used together with the U-ANT analyzer is used), the IX2-MLWPO DIC polarizer unit for MLWCD, and an applicable objective. 1 DIC Prisms and Applicable Objectives UIS2 Series DIC Prism Index Name Applicable Objectives IX2-MDIC20 MDIC20 UPlanSApo20X, UPlanSApo20XO, LUCPlanFLN20X, UPlanFLN20X IX2-MDIC40 MDIC40 UPlanSApo40X, LUCPlanFLN40X, UPlanFLN40X, UPlanFLN40XO (U-DICTS only, BFP-1 position used) UIS Series (Note) Usable regardless of the model number (3, 2 or none). 16 Fig. 7 2 Attaching the DIC Prism (Fig. 7) }The DIC prism can be attached in the same was as a modulator (see page 5). However, as the DIC prism has a positioning pin, it is required to align it with the groove. Align the on the DIC prism with the groove ² on the turret position and insert the DIC prism. # Be careful not to touch the DIC prism inside the frame. # Do not tighten the centering adjustment screw too much, for this may deform the DIC prism. 3 Attaching the IX2-MLWPO Polarizer Unit }See page 6. 4 Attaching the U-DICTS or U-DICT DIC Slider }The U-DICTS should be used in combination with the IX2-AN analyzer and the U-DICT should be used in combination with the U-ANT analyzer. 5 Attaching the IX2-AN or U-ANT Analyzer }Attach the DIC slider and analyzer on the IX2 series (IX81/IX7/IX51) or IX series (IX70/IX50) microscope frame by referring to its instruction manual. 13

17 IX2-MLWCD ² 6 Cross-Nicol Adjustment (Figs. 8 and 9) ³ Fig. }In the adjustment stage, it is recommended to use a general dyed specimen in order to facilitate the focus adjustment. If such a specimen is not available, use dirt or scratch on the petri dish for focusing. 1. Rotate the condenser for the BF light path. 2. Push in the slide lever ² of the IX2-MLWPO polarizer unit to engage the polarizer in the light path. 3. Engage the 10X or 20X objective in the light path and bring the specimen into focus 4. After obtaining the focus, remove the specimen, remove an eyepiece and look into the eyepiece sleeve to view the interference stripe. }The interference stripe can be magnified as described below. When the U-BI90CT binocular observation tube is used, rotate the CT turret to position CT to engage the CT lens in the light path. When the U-BI90 or U-TBI90 binocular observation tube or U-TR30H trinocular observation tube is used, remove an eyepiece and attach the U-CT30 centering telescope. Now bring the interference stripe into focus in the following method. U-BI90CT: Turn the focus adjustment ring. U-CT30: Turn the upper adjustment ring. 14

18 5. Rotate the prism movement knob of the DIC slider in the clockwise direction all the way until the knob is stopped. A black interference stripe then a rainbow-colored interference stripe will be observed. Here, stop the knob at the position with which the black interference stripe can be seen. (Fig. 9) 6. While observing the interference stripe, rotate the polarizer rotation screw ³ horizontally until the black interference stripe becomes darkest. Now turn the polarizer clamping screw ³ clockwise to clamp the polarizer there. Fig. 9 15

19 IX2-MLWCD 7 Observation Procedure }Set the prism position selector lever of the U-DICTS DIC slider to the pulled-out (NORMAL) position. 1. Rotate the condenser turret to engage the DIC prism matching the objective to be uses in the light path. 2. Engage the objective in the light path. 3. Place the specimen on the stage and bring the specimen into focus by moving the objective up or down. 4. Adjust the field iris diaphragm (FS lever) so that its image circumscribes the field of view. }If the field iris diaphragm has not been adjusted, adjust it by referring to the instruction manual for the microscope frame. 5. Stop down the aperture iris diaphragm (AS lever) to enhance the contrast. 6. Rotate the prism movement knob of the DIC slider to select the interference color that can provide the optimum contrast in accordance with the specimen. The background interference color is continuously variable from the gray sensitive color to purple sensitive color. Setting the background color to dark enables an observation like darkfield observation. Setting the background color to gray provides observation with high contrast and 3D feeling with the gray sensitive color with which the sensitivity is highest. There is a directional characteristic with the detection sensitivity because of the configuration of the DIC prism. As a result, the contrast may sometimes be improved by rotating the specimen on the stage. 16

21 IX2-MLWCD 2 Attaching the Phase Contrast Ring Slit }Attach in the same way as the modulator (see page 5). 3 Centering the Phase Contrast Ring Slit, Observation (Fig. 10) }Open the aperture iris diaphragm because stopping it down causes flare at the center. 1. Attach a phase contrast objective and bring the specimen into focus. 2. Engage the condenser s ring slit matching the phase contrast objective in use. }When the U-BI90CT binocular observation tube is used, rotate the CT turret to position CT to engage the CT lens in the light path. When the U-BI90 or U-TBI90 binocular observation tube or U-TR30H trinocular observation tube is used, remove an eyepiece and attach the U-CT30 centering telescope. Now bring the ring slit into focus in the following method. U-BI90CT: Turn the focus adjustment ring. U-CT30: Turn the upper adjustment ring. 18

22 Fig Insert the two centering knobs into the two centering knob insertion holes (marked ) and turn the knobs so that the ring slit overlaps with the phase plate ² of the objective. }When ghost images of ring slit image are visible, overlap the brightest image with the phase plate. }When the specimen is thick and it is moved or replaced, the overlapping of the ring slit and phase plate may be deviated and the observed image contrast become poor. In this case, readjust the centering by performing steps 1 to After completing centering, rotate the CT turret to position 0. If the centering telescope is in use, replace it with the eyepiece. }If the container is not flat, the second centering may be required to obtain the best contrast. In this case, begin centering with the lowest-magnification objective and increase the objective magnification in every centering. 5. Adjust the field iris diaphragm (FS lever) so that its image circumscribes the field of view, and begin phase contrast observation. }If the field iris diaphragm has not been adjusted, adjust it by referring to the instruction manual for the microscope frame. }The contrast may be improved by engaging the green filter in the light path. 19

23 IX2-MLWCD 3-5 Simplified Polarized Light Observation }Simplified polarized light observation requires an analyzer and polarizer. 1 Attaching the Analyzer and Polarizer 1. Attach the U-ANT or IX2-AN analyzer on the microscope frame. (For the assembly method, refer to the instruction manual for the IX2 series (IX81/ IX71/IX51) or IX series (IX70/IX50) microscope frame. 2. The polarizer should be the dedicated polarizer provided with the condenser or the IX2-MLWPO polarizer unit. Note that the IX2-MLWPO offers a higher performance. 2 Observation Procedure 1. Engage the analyzer and polarizer in the light path, place the specimen on the stage and bring it into focus. Now polarized light observation can be started. 2. Adjust the field iris diaphragm (FS lever) so that its image circumscribes the field of view. 3. Stop down the aperture iris diaphragm (AS lever) to enhance the contrast. 20

24 TROUBLESHOOTING GUIDE Under certain conditions, performance of the microscope may be adversely affected by factors other than defects. If problems occur, please review the following list and take remedial action as needed. If you cannot solve the problem after checking the entire list, please contact your local Olympus representative for assistance. Problem Cause Remedy Page a. The brightfield image glares and has poor resolution. The aperture iris diaphragm is stopped down. Open the aperture iris diaphragm. 3/8 b. The phase contrast ring slit does not match. An incompatible optical element is engaged in the light path. Select the optical element turret to match the objective. 17 An incompatible objective is engaged in the light path. Engage the optimum objective in the light path. 17 c. The polarization performance is poor. The polarizer is not engaged in the light path. Engage the polarizer in the light path. 11/15 The analyzer is not engaged in the light path. Engage the analyzer in the light path. 13/20 An optical element is engaged in the light path. Select the turret position without the optical element. The aperture iris diaphragm is opened. Stop down the aperture iris diaphragm optimally. 3/16/20 d. The interference color is not observed in DIC observation. The polarizer is not engaged in the light path. Engage the polarizer in the light path The analyzer is not engaged in the light path. In OLRC observation, the specimen cannot be observed with 3D feeling or the contrast is poor. An incompatible optical element is engaged in the light path.
