1 INSTRUCTIONS CX-PCD PHASE CONTRAST TURRET CONDENSER This instruction manual is for the Olympus Phase Contrast Turret Condenser Model CX-PCD. To obtain optimum performance and to familiarize yourself fully with the use of this condenser, we recommend that you study this manual thoroughly before operating the equipment. Retain this instruction manual in an easily accessible place near the work desk for future reference. A X

3 IMPORTANT CX-PCD This unit employs a UIS2/UIS (universal infinity system) optical design, and should only be used with UIS2/UIS microscope frames (CX40, CX41), eyepieces, objectives, etc. Less than optimum performance may result if inappropriate accessories are used. CAUTION If the condenser is operated in a manner not specified by this manual, the safety of the user may be imperiled. In addition, the equipment may also be damaged. Always operate the equipment as outlined in this instruction manual. FEATURES 1. The CX-PCD has 3 built-in ring slits that can be selected to allow phase contrast observation at 10X, 20X, 40X and 100X. Darkfield observation (10X or higher) and brightfield observation are also possible. 2. Each ring slit can be centered. 3. Compatible phase contrast objectives are: UIS2 series PlanCN 10Xph, 20Xph, 40Xph, 100XOph PlanN 10Xph, 20Xph, 40Xph, 100XOph UIS series Ach 10Xph, 20Xph, 40Xph, 100XOph Plan 10Xph, 20Xph, 40Xph, 100XOph 1

5 3 ASSEMBLY CX-PCD ³ ƒ 1 Mounting the Condenser (Fig. 1) ² Fig. # To secure space beneath the stage, remove the compensating lens. 1. Turn the coarse adjustment to raise the stage to its highest position. 2. Turn the condenser height adjustment knob ² to lower the condenser holder to the position where the clamping screw ³ can be turned. 3. Positioning the phase contrast turret condenser with the engraving at the front, insert the condenser into the mounting collar ƒ. Place the auxiliary knob over the clamping screw and tighten. After tightening, remove the auxiliary knob. 4. Turn the condenser height adjustment knob ² to raise the condenser holder to its highest position. 3
6 @ ² 2 Mounting the Compensating Lens (Fig. 2) }Use the compensation lens provided with the phase contrast turret condenser. 1. Insert the compensating into the condenser from below until it seats with click. 2. Turn the compensating lens so that the centering knobs ² are at the front. 7 4 PHASE CONTRAST OBSERVATION CX-PCD Centering the Condenser 1. Rotate the turret to the 0 index position and fully open the aperture iris diaphragm. 2. Place the specimen. Engage the 10X objective and bring the specimen into focus. 3. Using the field iris diaphragm knob on the microscope base, stop down the field iris diaphragm. 4. While looking through the eyepieces, turn the condenser height adjustment knob to bring the field iris diaphragm image into focus. 5. Rotate the two compensation lens centering knobs, shown in Fig. 2, to adjust so that the field iris diaphragm image is centered in the field of view. (Figs. 2, 3) To check centration, gradually open the field iris diaphragm. The condenser is properly centered if the field iris diaphragm image is precisely inscribed in the field of view. }While opening the field iris diaphragm, the iris diaphragm image may go out of focus. In this case, refocus it by using the condenser height adjustment knob. Fig. 3 When used for actual observation, open the field iris diaphragm slightly so that its image just circumscribes the field of view. 5
8 Centering the Ring Slits (Phase Contrast Observation) 1. Engage the phase contrast objective to be used. 2. Rotate the phase contrast turret to match the Ph indication with the Ph indication of the phase contrast objective. Ex.: If the Ph2 is engraved on the objective, Ph2 should also be seen in the window on the turret. 3. Place the specimen and bring it into approximate focus. 4. Remove one of the eyepieces and insert the focusing telescope (U-CT30) into the eyepiece sleeve. 5. Rotate the knurled ring of the U-CT30 to bring the bright ring (condenser ring slit) and the dark ring (objective phase annulus) into focus. 6. While pushing slightly inward, rotate the two ring slit centering on the turret condenser until the bright ring and the dark ring are concentric and superimposed. 9 CX-PCD The ring slits corresponding to the other objectives should be centered in the same manner. In case Ph1 is indicated on both 10X and 20X objectives, for observation purposes, once the ring slit is centered for the 10X, it is unnecessary to re-center it for 20X. However, for the purpose of photomicrography, it is recommended the ring slit be centered for each objective even if the same Ph number is indicated on the objective. 7. Remove the U-CT30 and replace with the eyepiece. Phase contrast observation can now be started by engaging any of the phase contrast objectives together with its corresponding ring slit. Darkfield Observation 1. Rotate the turret until the DF indication is shown. 2. Engage the 10X or higher objective. # Objectives with NA 0.7 or lower NA and objectives with iris diaphragm can be used. 3. Place the specimen on the stage and bring the specimen into focus. 4. Adjust the height of the condenser until a darkfield field of view with uniform illumination is obtained. 5. Open the field iris diaphragm to the extent that even brightness is obtained. # To avoid any glaring effect, keep eyes away from the eyepieces when changing the objective during darkfield observation, or when changing from darkfield mode to another observation mode. 7
