3 SAFETY WARNING KEEP HANDS AND CLOTHING CLEAR OF ANY MOVING PARTS WHEN IN OPERATION DEFINITIONS: Home Z Moves the Pin Tool to the top of the Lead Screw range, reads a sensor and sets the Z motor counter to Home Carousel Moves the Carousel to Deck Station #1. Sets the counter for the rotational Servomotor to The Carousel will reposition itself to Station #16 to get off the Station #1 homing sensor. Max Z The Z height that will allow clearance of the tallest reservoir or obstacle on the deck. This height must be reached before the rotational motor can advance. After every Z movement in a Mother, Daughter, Blot, Wash, Auxiliary A or Auxiliary B station the Pin Tool moves up to the Max Z position prior to rotation. Page 3

4 STEP BY STEP OPERATING INSTRUCTIONS 1. Remove top and sides from shipping crate. Grasp the Pin Tool Robot under the bottom plate and lift on to a counter top or bench (3 to 4 person job). INITIAL SETUP 2. Use adjustable legs to level the Rotary Deck. Make sure that Pin Tool Robot is not resting on rotary motor that extends below the bottom plate. 3. Plug in both power cords and turn on the main power switch on Touch Screen. Main Power Switch 4. Touch Home Z on the V&P Scientific screen. The Pin Tool Positioning Jig will move to the top of the Lead Screw, and home to the Z Sensor and stop. While it is homing the screen will flash Homing. When the screen stops flashing the Positioning Jig is Homed and will stop. Page 4

5 INSTALLING PIN TOOL 1. The Back Positioning Screws and the Left Positioning Screws have been backed off prior to shipment to facilitate the installation of the Pin Tool. Use a 1/16 Allen wrench (provided) to loosen the nylon Locking Set Screws. Front Swivels Locking Set Screws 2. Lift the two Front Swivels up to allow the Mounting Plate to slide into the Jig and be located between the Positioning Screws. Note: The Front Swivel Positioning Screws should be kept in the down or lowered position at all times other than loading or unloading the Pin Tool. The Pin Tool Jig will jam if the Swivel Positioning Screws are up during Home Z operations. Swivel Positioning Screw Page 5

6 3. Seat the Pin Tool by lowering the Mounting Plate of the Pin Tool in the Jig against the left side and then lower the right side into the jig. Use a 3/32 Allen wrench (provided) to adjust the Left and Back Positioning screws so they push the mounting plate against the Front and Right Side Positioning Screws. Left Side Positioning Screws Mounting Plate 4. Once the Left and Back Positioning Screws are tight, check to see if the Mounting Plate is securely held in the jig by wiggling it. Repeat until it does not wiggle. Mounting Plate Page 6

7 ADJUST PIN TOOL ALIGNMENT POSITIONS 1. The Pin Tool can be precisely adjusted in the X and Y orientation as well as rotationally by adjusting the eight Positioning Screws. Loosening or tightening the two Left Side Positioning Screws on the Jig adjusts the X position. The Y position is adjusted using the two Back Positioning Screws. See photo and instructions below before adjusting. Back Positioning Screw Positioning Screw on Front Swivel Left Side Positioning Screw 2. The Front Swivel and Right Positioning Screws are set at the factory and fixed in place with Locking Set Screws. They should not need adjusting. However if they do, be sure to loosen the Locking Set Screws. Loosen the nylon Locking Set Screws (1/16 Allen wrench) on the swivels prior to adjusting the Y Positioning Screws on the Front Swivels. 8 3. Nylon Locking Set Screws for the Right Side Positioning Screws, the Left Side Position Screws (X axis) and the Back Positioning Screws (Y axis) are located on the bottom of the Jig. Loosen all Locking Set Screws prior to adjusting the Positioning Screws. Locking Set Screws 4. Two slot shaped holes (arrows) are in the lower Z superstructure to provide Allen wrench access to adjust the Right Side Positioning Screws. There are also two holes near the top of the superstructure that can also be used when Jig is in the Home Z position. 5. At the initial installation and at times during use, you will need to determine if positioning adjustments need to be made to the Pin Tool Jig. Touch Home Carousel on the V&P Scientific screen (red arrow). The Robot will home the Carousel to Station #1 and then reposition itself to Station #16. The Homing Complete - OK to Continue screen will flash when the Home Carousel operation is complete. Page 8

9 6. To determine the Positional Alignment of the Pin Tool: Place a VP 903R-96, VP 903R-384 or a VP 903R-1536 Registration Standard (corresponding to your Pin Tool) into Carousel Station #1 (or any position of your choosing). Then perform the follow steps to lower the Pin Tool to the Registration Standard Touch Index Carousel (see screen in Step 4 for location of Index Carousel button). 8. Touch New Station (red arrow). Input 1 then select the Return Key. 9. Touch Begin Carousel Move (green arrow). The Carousel will move to Station # When the Carousel stops at Station #1, touch Back to get to the Main Screen, then touch Setup. Page 9

10 11. Touch Teach Positions on the Setup Screen. 12. Touch Mother Plate on the Teach Positions screen. 13. Touch the Jog down arrow until the four corner pins of the Pin Tool almost touch the four corner spots on the Registration Standard. Note: Use care in Jogging down as the pin tool will move quickly and takes a while to decelerate. 14. Adjust the Fixed Positioning Screws (steps 1-4) in the Jig until the corner pins line up with the corner registration spots. Page 10

11 15. Touch the Jog up arrow until the Pin Tool Jig is near the Home Z Height. This will raise the Pin Tool for easy access to the nylon Locking Set Screws. 16. Tighten the eight nylon Locking Set Screws. Confirm the Pin Tool alignment with the Registration Plate by touching the Jog down arrow until the alignment of the pins to the Registration Spots can be verified. Once the alignment is confirmed, touch the Jog up arrow until the Registration Plate can safely be removed from Carousel Station #1. Locking Set Screws 17. Touch the Back on Mother Plate Setup Page 1 screen (see step 14 above) to get to the Teach Positions screen. Touch the Max Values. Proceed with step #2 of the SETTING MAX Z VALUES TO PREVENT PIN TOOL CRASHES section. Page 11

12 SETTING MAX Z VALUES TO PREVENT PIN TOOL CRASHES 1. From the Setup Screen touch the Teach Positions, which will take you to the Teach Positions screen. Touch Max Values (red arrow). NOTE: THIS STEP IS VERY IMPORTANT IN PREVENTING PIN TOOL CRASHES 2. Touch Security Login on the Max Height Setup screen (red arrow). 3. This will bring you to the Password screen. The password set at the factory is numeric. Touch the 0-9 key (red arrow), then the Enter key (blue arrow) to switch the display from the alphabet to numeric. Page 12

13 4. The password is Touch those number keys (red arrow) and the Enter key (blue arrow) to register the password. 5. Touch the Jog up or down arrow until the Pin Tool clears the tallest obstacle on the carousel (red arrows). Touch the Push to Set Button (blue arrow) and verify that the Current Position and the Preset Position are the same (purple boxes). 6. When pressing Push to Set in Step 5, a second screen is displayed: WARNING 1234 and Y and N. Touch the Y (red arrow). You will then be allowed to set the Max Z Height. Touch Next (blue arrow) to advance to the Default Speeds Setup screen. Page 13

14 7. Touch the Max Z Speed indicator at the % you want to use. We recommend 100%. The Max Z Speed refers to the Z Speed from the Z Height of any plate (Mother, Daughter, etc.) to the Max Z Height. The Z Axis Home speed refers to the Z Speed from Z Max to Z Home. The Table Run refers to the speed that the carousel spins. All three of these speeds are controlled by touching the indicator screen at the desired % or by sliding your finger to the desired setting. 8. When setting the Default Speeds, the second screen appears displaying: WARNING 1234 and Y and N. Touch the Y (red arrow). You will then be allowed to change the Max Z Speed. 9. Set the Z Axis Home and Table Run speed scales as described in step 7. These can be set as high as 100% if you choose. (At 100%, liquids will not slosh out of baffled reservoirs such as VP 540L with VP 540LF baffles). 10. Touch Back (blue arrow) and return to the Teach Positions screen. Your speeds will be recorded and saved. Page 14

15 TEACH POSTIONS: Z HEIGHTS, CYCLES, PECK DELAY AND SPEEDS 1. From the Teach Positions screen touch Mother Plate. 2. Place a dry Mother Plate in Carousel Position #1. Touch the Jog down arrow until the pins are near the bottom of the plate (red arrow). Advance slowly to the point where the pins just begin to rise as they touch the bottom of the plate. Touch the lower Push to Set button (blue arrow) and verify that the number in the Current Position (orange box) and the lower Preset Position (blue box) are the same. Next, touch the Jog up arrow until the pins just clear where the liquid level will be (green arrow). Touch the upper Push to Set button (purple arrow) and verify that the number in the Current Position (orange box) and the upper Preset Position (purple box) are the same. 3. Touch Next to got to Page 2. Page 15

16 4. Adjust the Pin Tool Z Speed by touching the Speed, Up % indicator bar at the % you desire. Repeat with the Speed, Down %. This controls the speed of dipping in and out with the pins (mixing). The Final % is the final Z Up Speed. Set the Final Z Up Speed as described above for the Up % and Down %. The Final Z Up Speed will determine how much liquid is carried on the pins. The faster the Z Up Speed, the more liquid that is transported. Use this Final Z Up Speed to adjust the volume transferred. 5. Touch Next (blue arrow) for Page To set the number of up and down Z Cycles that the Pin Tool will perform, touch the # of Deck Cycles (red arrow). This will bring up a numeric screen seen below. 7. The Peck Delay time is also set on this screen (blue arrow). The Peck Delay is how long the Pin Tool remains at the bottom of the down Z Cycle. For mechanical reasons, we recommend that this number should always be above 30, which is 0.3 seconds. 8. Touch the appropriate number to enter the desired number of Deck up and down Z Cycles or to set the Peck Delay (green arrows). 9. Touch Teach New Plate to continue with teaching different plates or Exit to Main for other functions. Page 16

17 10. Follow steps 1 to 8 for each of the rest of the plate definitions: Daughter Plates, Wash, Blot Plates, Auxiliary Plate A and Auxiliary Plate B, and *Wash Plates. We recommend at least 3 dipping cycles for Wash Plates and Daughter Plates. We also recommend a 3 second Peck Delay on the Blotting Plate to allow liquid to drain off the pin. One deck cycle is sufficient for blotting (0 cycles). 11. Touch Exit to Main when complete or go to Teach Wash Plates Positions screen by touching Wash Plates (red arrow) to. * Wash Plates require an extra screen shown below. 12. Teach Wash Plate Positions in this screen refers to teaching different positions for use in the Wash Cycle. The Wash Cycle routine is used to clean the Pin Tool at the beginning and end of a Regular Cycle routine (see also page 29). 13. Follow same steps 1 to 8 as above to teach these positions. Page 17

18 SETTING STATION OFFSETS FOR PIN TOOL ROBOT 1. From the Setup Screen touch the Teach Positions, which will take you to the Teach Positions screen. Touch Station Offset (red arrow). Note: This has been done at the factory and it is not recommended that the user re-adjust these settings. If it does become necessary please call V&P Technical Service at for assistance. 2. Touch Security Login (red arrow). This is a second level Security Login that allows for these settings to be protected from being accidentally changed during everyday use. This is a separate Security Login from that used in setting Max Z and Speed parameters in Teach Positions section. 3. This will bring you to the Password screen. Call V&P Technical Service at to obtain the password set at the factory. Touch the keys for the password (red arrow), then the Enter key (blue arrow). Page 18

19 11. Touch a Station Offset position to be adjusted (red arrow) and a second screen is displayed: WARNING 1234 and Y and N. Touch the Y (blue arrow). 14. Touch the appropriate number to enter the desired distance of offset for the selected station (green arrows). 15. Confirm alignment as before in ADJUST PIN TOOL ALIGNMENT POSITION section by lowering the Pin Tool to the Registration Standard (page 7). Page 19

20 1. Touch Configure Robot Setting on the Setup Screen. CONFIGURE PLATE HANDLING ROBOT 2. Each Station of the Plate Handling Robot needs to be taught: the Robot Plate Stacks 1 to 5 and the position to load plates onto the Pin Tool Robot Deck or P. Touch the first Station to be taught, This screen allows you to set the rotary position for the chosen Robot Station. The Current Quantity is the number of plates that the robot last counted in the position (red arrow). It will be updated when it next tries to pick up a plate. If a Preset Position exists, touch the Go button to move the robot to that position (blue arrow). Using the Jog Arrows adjust the position of the Robot Gripper Arm to align down the middle of the Stack (green arrow). See photo on next page. When the position is correct, touch the Push to Set button (purple arrow) and Page 20

21 the Preset Position and Current Position should now match. Touch Back to return to the Robot Settings screen. Repeat for each Robot Station. 4. Next is the Pin Tool Robot Deck or P position. The first step is to load a plate in the Robot Gripper. Do this by placing a plate in any stack. Touch the 1-5 button that corresponds to the stack where the plate was placed (#1 is shown as an example). 5. The appropriate Robot Station screen will open and will have the Preset Position showing the correct position. Touch Go (red arrow) and the Robot will position itself over the correct stack. Then touch Back to go to the Robot Settings screen (blue arrow). Page 21

22 6. The Robot Settings screen will appear. Touch Robot Operations. 7. Touch Find Plate (red arrow), the Robot will move downward until it senses a plate. It will stop. Touch Close Gripper (blue arrow). Touch the Home Vertical button (green arrow) and the robot arm will raise the plate above the stacks. Touch Back to return the Robot Settings screen (purple arrow). 8. Touch the P button to open the Pin Tool Station Screen. Page 22

23 9. The Pin Tool Station requires both a rotary and a vertical setting. If there is a Preset Position in the rotary Preset Position, touch Go to move to that position (red arrow). Use the Vertical Jog (blue arrow) to lower the plate to a height just slightly above the Plate Holding Pins of the Pin Tool Robot Carousel Station to be taught. Using the Rotary Jog (green arrow) and by physically moving the Plate Handling Robot and the Pin Tool Robot, line the two machines up so that the plate will just clear the Plate Holding Pins in the Carousel Station. Use the Vertical Jog (blue arrow) to lower the plate to the surface of the Pin Tool Station. Again adjust the position of the two robots to align the plate in between the Plate Holding Pins. Plate Holding Pins When the location is correct, touch the Push to Set buttons in both areas (purple arrows) and verify that the preset position and current position are the same. Then touch Back to return to the Robot Setting screen and then touch Back again to return to the Setup Screen. Note: Make sure the Plate Handling Robot rotary arm also vertically clears plates or other items in Pin Tool Robot Carousel Stations adjacent to the one being loaded. Page 23

24 Note: The spring-loaded Plate Holding Clip that holds the plate tightly into the Plate Holding Pins can also be adjusted. Place plate or other accessory into Deck Station where it will be used and adjust fit with set screw located in the side of the Clip. Page 24

25 1. Touch Configure Regular Cycle in the Setup Screen. CONFIGURE REGULAR CYCLE 2. Touch Configure Plates in Setup /Configuration screen. 3. This screen allows you to designate which of the six different Plate Definitions is in each Station on the Pin Tool Robot Carousel Deck (Mother, Daughter, Blot, Wash, Plate A, Plate B). It also allows you to designate which Carousel Stations are empty. Page 25

26 4. Touch a Carousel Station #, from 1-8 (red arrow). A second screen overlays the Station#/Plate Type screen. Select the type of plate that will be used in that station by touching the corresponding button (blue arrow). Up and down arrows allow you to move up and down the plate type list. Touch Next (green arrow) and it will advance to stations From the Stations 9-16 screen touch CFG Main button to return to the Setup/Configuration screen. 6. Touch Configure Routine. This will allow you determine the flow of the Pin Tool Assay Routine. Page 26

27 7. The Assay Routine for the VP 903B can accommodate 256 steps. This allows for some stations such as Mother Plates, Blotting Plates and Wash Plates to be used multiple times in a single assay protocol or routine. 8. Touch a Routine Layout Order square (red arrow) to place the Carousel Station number into the routine layout. Touch the appropriate numbers (blue arrow) followed by return key (green arrow) to input the desired Robot Move using the rules described below. 9. For Pin Tool Robot only moves, the first two digits must be 0. The last two digits can be the 00 to 16. So for all operations for the Pin Tool Robot the numbers should be 0001 to An entry of 0000 means no operation or End of routine. 10. Moves involving the Plate Handler Robot have a different format: The first two digits 0 to 16 signify what Station on the Pin Tool Deck the plate is either to be placed in or retrieved from. The next two digits determine the type of Plate Handler move. A 1 to 5 in the third digit, followed by a 0 in the fourth digit signifies that the Plate Handler Robot is to remove a Pin Tool Station 11 move, no Plate Handler Robot: Pintool Robot Only Plate Handler Robot moves plate from Stack 2 to Pin Tool Station 11: Pintool Robot Plate Robot Page 27

28 plate from the Stack specified in the third digit and place in the Pin Tool Station specified in the first two digits. For example 1120 means the Plate Handler Robot takes the top plate from Stack two and places in Station 11 on the Pin Tool Robot Deck. To reverse the operation, the fourth digit is used instead of the third digit means the Plate Handler Robot removes the plate at Pin Tool Station 11 and places on top of Stack two. Plate Handler Robot moves plate from Pin Tool Station 11 to Stack 2: Pintool Robot Plate Robot Continue placing Carousel Station numbers in the Routine Order until you reach the end of the routine. Place 0000 in that space and touch Back to go to the Setup/Configuration screen. 12. Touch Configure Cycles to go to the Routine Cycles screen. Page 28

29 13. Set the number of cycles you want to run a particular routine by touching the number box screen. 14. Touch the number on the screen and then the return key. Touch Back until the Setup Screen is reached. Page 29

30 1. The Wash Cycle is used to clean the Pin Tool at the beginning and end of a regular cycle run. Touch Configure Wash Cycle. CONFIGURE WASH CYCLE 2. Touch Configure Wash Plates. 3. Touch one of the Wash Station positions to configure the Wash Plate Type at that station. Page 30

31 4. Use arrows to jog up and down (red Arrow) and select desired plate type (blue arrow). Touch Back (green arrow) to return to Setup / Configuration screen. 5. Touch Configure Wash Routine. 6. Touch the square in the Layout to enter the Station # corresponding to the step in the Wash Routine. Page 31

32 7. Touch the appropriate number to enter the desired Station number. Touch Back until you get to the Setup Screen. Page 32

33 1. Touch Back until you arrive at the Main Screen, then touch Sys Ready Run. RUNNING A CYCLE 2. Touch the desired type of Run Cycle. The following screens and information pertain to both types. 3. When the Pin Tool Robot is running a routine, it displays on the Run Screen the current Carousel Station running, the # of Cycles completed and how many total Cycles are in the routine. When the Plate Handler Robot is moving Robot Mode flashes. Page 33

34 1. To stop the routine at any step, touch Running (red arrow). STOPPING AND RESTARTING A CYCLE 2. To restart the routine where it was paused, touch Stopped (green arrow). 3. To restart at the beginning of a cycle, touch Max Z Position (red arrow). 4. Then touch Max Z Position Restart Cycle (green arrow). Page 34

35 Note: Safety feature in software will stop the program if the Z motor encounters significant resistance in its downward movement. When this happens the screen at the right will appear. Turn the power off and back on again, then determine and correct the problem. Page 35

36 Important Setup Notes: CLEANING PIN TOOL It is best to add the liquid to the reservoir after the reservoir is placed on the carousel to avoid spills. It is also best to remove the liquid from wash reservoirs before removing them from the carousel. The Pin Cleaning Solution is a weak acid but will remove the anodized layer from the aluminum carousel. Clean up spills immediately. The volume in the 3 wash reservoirs is increased from 120 ml of Pin Cleaning Solution to 130 ml of water to 140 ml of alcohol. This is to insure that a compound high water mark ring does not form on the pins. Use care to determine that each plate is pushed into the left corner for proper registration with the spring loaded arm. 1. The following cleaning protocol will run on the Pin Tool Robot if the Wash Cycle protocol has been previously entered with the following assignments: Carousel Stations 4, 6 and 8 have VP 540L-1536 Wash Reservoirs and Carousel Stations 5, 7 and 9 have Blot Pads. 2. Turn Touch Screen power switch off and then on again. 3. Touch Home Z on the opening screen. 4. Place a baffled VP 540L-1536 Reservoir with 120 ml of VP 110 Pin Cleaning Solution (diluted 1:5) in Carousel Station #4. 5. Place a VP 540DB Blotting Pad in Carousel Stations #5, 7 and 9. VP 540DB VP 540L-1536 with VP 540LF Place a baffled VP 540L-1536 Reservoir with 130 ml of distilled water in Carousel Station #6. 7. Place a baffled VP 540L-1536 Reservoir with 140 ml of methanol, ethanol or isopropyl alcohol in Carousel Station #8. VP 540DB Page 36

37 8. Remove all plates from Carousel Stations 1, 2 and Touch Home Carousel (red arrow). The Robot will home the Carousel to Station #1 and then reposition itself to Station #16. NOTE: The Front Swivel Positioning Screws should be kept in the down or lowered position at all times (unless loading or unloading the Pin Tool.) The Pin Tool Jig will jam if the Swivel Positioning Screws are up during Home Z operations. 10. Touch Homing Complete-OK To Continue (blue arrow) to go to Main Screen. 11. Touch Sys Ready Run to go to Run Screen. Page 37

Touch Run (red arrow) and the Pin Tool Robot will start Wash Cycle Routine as described in the RUNNING A CYCLE on page Upon completion replace the wash reservoirs with the wash liquids called for in the assay process. The VP 540DB Blot Pads can remain. For example, place a Mother Plate in Carousel Station #1, a Daughter Plate in Carousel Station #2 and a VP 540DB Blot Pad in Carousel Station #3 (see photo previous page). 14. Touch Run Cycle (red arrow) to run an assay routine. Page 38

39 1. Place assay plates, wash reservoirs and blotting plates on carousel deck stations to correspond with configured routine. Note: Use care to determine that each plate is pushed into the left corner for proper registration with the spring loaded arm. 2. We recommend that you fill the wash reservoirs while on the deck: fill the first wash reservoir with 120 ml wash liquid (liquid type depends on the sample being transferred), the second with 130 ml of water and the alcohol reservoir with 140 ml. This is to insure that a compound high water mark ring does not form on the pins. Depending on how many cycles are run and the amount of time involved, you may want to add liquid to the reservoirs especially the alcohol reservoir, as it will evaporate at a faster rate. 3. Turn power switch off and then on then touch Home Z. NOTE: The Front Swivel Positioning Screws should be kept in the down or lowered position at all times other than when loading or unloading the Pin Tool from the Jig. The Pin Tool Jig will jam if the Swivel Positioning Screws are up during Home Z operations. RUN ROUTINE Page 39

40 4. Touch Home Carousel on the V&P Scientific screen (red arrow). The Pin Tool Robot will home the Carousel to Station #1 and then reposition itself to Station # The Homing Complete - OK to Continue screen will flash when the Home Carousel operation is complete. 6. Touch Homing Complete-OK To Continue to go to Main Screen (green arrow). 7. Touch Sys Ready Run to go to Run Screen. 8. Touch Run and the Pin Tool Robot will execute the routine.
