2 Abstract When large biomolecules such as IgM are purified, group separation with gel filtration is a technique typically used as the polishing step to secure high purity. The drawback of gel filtration is the relatively low productivity. Capto Core 700, a novel chromatography medium (resin) from GE Healthcare, designed with core bead technology, shows promising results for high productivity group separation in the of IgM 2

3 Abstract The core bead technology enables double functionality in Capto Core size exclusion and binding separation in one bead. The benefits of this technology include high loading, high flow rates with short residence times, and robust performance in a wide window of operations, securing high productivity and straightforward process design. 3

4 Layered beads Layered beads are novel type of chromatography bead from GE Healthcare. Traditional chromatography beads from GE Healthcare can be described as hydrophilic, porous beads homogenously functionalized with ligands. With layered beads, GE Healthcare has developed a technology to functionalize the beads in different layers. When designing a layered bead there are a number of new design variables to keep in mind. Depending on how these design variables are chosen (Fig 1), different and interesting chromatography properties and features can be introduced. 4

6 Core beads One type of layered beads is core beads. In core beads the ligands are situated exclusively in the core (interior) and the outer layer is inactive (not functionalized with any ligands). A core bead design enables the bead to offer double functionality. Size exclusion (from the outer layer) and binding separation (through the ligands) in the core of the bead. 6

7 Core beads The cutoff is defined by the pore size in the outer layer, while the binding separation is defined by the ligand used in the core of the bead. Depending on the size of the target a core bead could be designed and optimized for either bindelute or flow-through mode (Fig 2). 7

9 Capto Core 700 Capto Core 700 is a chromatography medium developed for of viruses and other large biomolecules. This medium is designed for group separation applications where the (large) targets go in the flow-through while smaller contaminants bind in the core of the beads. 9

10 Capto Core 700 Capto Core 700 has an inactive layer with a cutoff of Mr ~ The ligand in the core is octylamine, which allows for an efficient binding of a wide variety of impurities. This medium offers high loading, high flow rates (Fig 3), and high yield and purity, leading to high productivity. Capto Core 700 has a wide window of operations allowing for straightforward process design. 10

12 High productivity of IgM When measuring the dynamic binding capacity (DBC) on Capto Core 700 for biomolecules with increasing molecular weight, there is a size related limit for DBC that is related to the cut off of the bead. As can been seen in Fig 4 the DBC for IgM (Mr of ~900 kda) is close to zero. This means that IgM is almost totally excluded from the interior of the bead and makes Capto Core 700 effective for of IgM. 12

14 High productivity of IgM In collaboration with Morphotek Inc., Capto Core 700 was evaluated for IgM. Initial screening showed that to secure high recovery, residence time was an important parameter to evaluate when working with a target molecule so close to the cut off of bead. As shown in Fig 5, there is a decrease in HCP removal with increased loading. At the same time, the recovered IgM increased with increasing loading. Thus the recovery of IgM to the HCP reduction needs to be optimized in order to secure the best performance 14

16 High productivity of IgM These initial screening experiments showed that Capto Core 700 could be a valuable tool in of IgM. Good levels of recovery, activity, purity and process time were demonstrated but further optimization of loading conditions (e.g., ph, conductivity, flow, concentration of load material) are suggested for performance improvement and to secure process robustness. 16

17 Conclusion Capto Core 700 has a molecular weight cutoff in the range of Mr making it an excellent choice for IgM. Optimizing residence time and loading conditions can lead to high IgM recovery, while reducing HCP levels. Capto Core 700 utilizes core bead technology to enable high loading and high flow rates, resulting in high productivity, group separation of large biomolecules. 17
