2 Wide range of products for numerous Applications AMS Biotechnology supplies a unique range of magnetic and nonmagnetic silica beads in the range from 150 nm up to 5 µm for customized beads and own bead production. Our standard products have sizes of 600 nm and 1 µm and 1.2 µm. For most applications the 1 µm beads are ideal. The 600 nm particles give more surface area per mg of bead but the magnetic strength slightly decreased. The 1.2 µm beads are used for genomics and proteomics applications where highest magnetic strength is needed. MagSiproteomics C8 beads represent an intermediate hydrophobicity (less hydrophobic than C18 and more hydrophobic than C4 beads) and are suitable for sample preparation in context to proteomic profiling and biomarker research. The relatively low hydrophobicity of the C4 beads allows the purification and fractionation of larger biomolecules like proteins. 5 1.3 MagSiSTA MagSiSTA beads are uniform silica particles with a monolayer of high quality streptavidin covalently attached to the bead surface. They come as readytousesystem. They can be used to isolate biotinylated proteins or other biotinylated molecules. The magnetic properties allow easy an quick washing steps in ELISA assays and protein or DNA/RNA isolations. In addition, since beads are in suspension and therefor the coupled antibody the incubation time can be shortened when compared to ELISA tests using antibody coated micro plates. 1.4 MagSiprotein A and G Protein A and protein G are recombinant proteins that bind with high affinity to the Fc portion of several classes and subclasses of immunoglobulin from a variety of species. Both proteins form reversible complexes with different IG subclasses and species. MagSiprotein A/G is is an ideal tool for efficient and economical purifications and immunoprecipitations from small sample volumes. The recombinant protein G (~26kDa) on the surface of our beads contain only IgG binding sites. The albumin, cell wall and cell membrane binding domains, present on the wildtype protein G, have been removed to optimize binding properties. The 42kDa protein A is from Staphylococcus aureus. The advantage of protein G an A beads, compared to Streptavidin beads, is that the antibody does not need to be biotinylated and the binding is reversible. So they are ideal for protein isolation and proteomics. Also, the magnetic properties allow easy and quick washing steps in protein isolations. 7 1.11 MagSiWCX and MagSiWAX MagSiWCX (weak cation exchange) and MagSiWAX (weak anion exchange) beads are an ideal tool for the reduction of protein or peptide complexity (e.g. cell lysates). Main applications include: Sample preparation and prefractionation prior to mass spectrometry (e.g. MALDITOF analysis) SDSPAGE analysis Protein and peptide separation for multiple downstream applications Biomarker analysis and serum/plasma profiling The high magnetic strength makes them applicable for both manual and automated/robotic fractionation, because the beads will typically collect in less than 1 minute when magnetic force is applied. This quick and complete separation gives very good reproducibility since no beads will be lost during washing steps. 8 Applications 2.1 Biocompatibility Different tests in blood and cell cultures show that MagSi beads have little to no toxicity. When incubating lung cells (A549) with regular MagSi silica beads, no increase in IL6 or IL8 was visible up to 500 µg/ml of beads. Comparable results were found for human whole blood. This means our beads are suitable for applications in living cells or blood when you want to keep the cells viable and the level of cell stress very low. 2.2 Cytokines in whole blood 0,8 Con IL6 and IL8 (ng/ml) 2. A biotinylated primary antibody is bound to the streptavidin to isolate the target protein, followed by detection of that protein with a labeled second antibody. OD 405nm 4 3 To measure IgG, a biotinylated antiigg is bound to the MagSiSAV bead. After incubation of the beads with an analyte, a second antibody labeled with peroxidase is added. Then substrate (ABTS*) is added and the green color development measured via a micro plate reader (absorption at 405nm, corrected for dilution). There is no difference visible between the MagSi beads (dashed line) and the reference beads (solid line) IgG (ng/ml) * 2,2 Azinobis(3ethylbenzothiazoline6sulfonic acid) When testing our streptavidin beads in an automated system (e.g. Bioveris MSERIES ) the noise level is very low compared to references and they have good reproducibility. 2.3 Minimal nonspecific absorption 1 The knowhow in our company and flexibility of production has allowed us to develop beads with low background absorption. When a protein needs to be isolated from a mix of other proteins, this background absorption determines the yield and purity of your target protein. MagSi bead tailored blocking methods reduces the nonspecific binding of proteins to an absolute minimum. In the image on the right side there is a SDS PAGE gel of MagSiSTA and MagSiprotein A/G beads after incubation with bovine IgG and BSA (fraction V) solution. For the protein G beads (lane 24 and 6), the IgG bands are clearly visible. The protein A bead (lane 5) doesn't show IgG, because the affinity for bovine IgG is very low. The streptavidin beads (lane 7) show no protein binding. No background absorption of BSA is visible for all the samples SDS PAGE after incubating beads in BSA solution. Lane 1: marker, lane 2 reference sample, lane 37 MagSi beads Downstream applications Tests show that MagSi beads don't interfere with downstream applications. When performing realtime PCR, there is no inhibition visible. Also when performing protein isolation, no artifacts occur during SDSPAGE or spectrometry. The same is true for biochemical assays like an ELISA.
9 2.5 Imaging Beads with a fluorescent dye or quantum dots make them perfectly visible under a fluorescent microscope. This means, specific cells can be visualized using fluorescent beads coated with an antibody (direct, via protein A/G or via streptavidin). In some cases, the cells can then be isolated by using the magnetic properties of the beads. < 4 samples of fluorescent labeled beads, a: Singel Quantum Dots, b: Quantum Dots in a vial, c: beads with rhodamin incapsulated, d: MagSi beads coated with single quantum dots 2.6 Nucleic acid isolation 1 The porous surface of the silica beads engineered for DNA/RNA isolation gives them a high surface area and high binding capacity. This makes sure that nearly all the DNA is isolated from the sample. The beads are ideal when using them to isolate DNA from cell lysates or whole blood. The high (up to 90%) concentration of ferro fluid makes it very easy to collect the beads, using a magnet, in the rinsing steps. Within seconds, all the beads are collected to the magnet. This high magnetic strength makes them also applicable in automated systems because it reduces loss of sample in the rinsing steps Agarose electrophoresis.
