6 Part I. Introduction and Summary 3. When power switch on?. Sign in SP-3000 DB supports Sign in mode for protecting personal information and information of personal measurement. Initial display after Self Test shows two Sign in kinds. One is Sign in and the other is Guest.. Sign in 3. When power switch on? When the power of the unit has switched on, the unit starts operating Self Test of status devices and defects of the unit. It shows results after it checks about each items. When Self Test, found defects on devices, and error message will apear as Error on the screen. The unit remains for confirmation of each Self Test. When the unit stays in good to process, Self Test display turns to the mail screen to process measurement and setting devices automatically. When an error/error appeared in Self Test display, contact After Service team(a/s) and technical assistance of Part III. When the power has been switched on, preheat the unit for more than 30 minutes is suggested to use with accurate results. [Sign in] : Personal account to Sign in that has been registered by one user. - Quick Measure : The function suggests to quick measure with previous measurement setting without input or setting automatically. - Protect individual measurement method, information data and more. [Guest] : Suggested new account to operate measurement by guest (Guest means the one who does not use constantly). 0

12 2-3. Quick Measure When the user has registered SP-3000 DB account previously, then you already have an account to proceed Sign in, touch [Sign in]. It will be also suggested to use when there are saved methods with registered measuring procedure. 2. When you select [Yes], go to measuring setup list. Select a method Click Load Go to the selected measuring mode Qcick Measure 3. When you select [No], go to set Quick measure mode (Condition already shows at the bottom of Quick measure button) Qcick Measure Quick Measure Mode. 13 4. In case of there are no saved method or no measuring procedure, the belowed message will pop up. Just after saving method or measuring procedure, using Quick measure mode possible. 3. Measurement setup To proceed measurement, setting condition must be saved as users necessity. SP-3000 DB suggests to proceed measuring simple wavelength measurement to know absorbance, transmittance and concentration, survey scanning, kinetic scanning, fast scanning in wavelength and time by operating Hybrid measurement mode. There are two sensors to manage operation, PD sensor and CCD sensor. How to save the method. Set up measuring condition with [Measure]- [Save setting], find [Save] on the bottom of display to proceed saving method. Setting devices to proceed a method. 3. Measurement setup 2-3. Qcick Measure Name Desicription 2. Input the name of the saved setting with touch [Save]. The saved method will be added to menu in [Quick measure]. Name Cell Type Unit Factor Memo Input your test name. Select preferred cell type (Refer to 6. Change and select cell type and holder). Select preferred units. Input factor for applying to dilution and test result. 14 Setup methods (abbreviations and information) PD Sensor / CCD Sensor 3. Measurement setup. Simple wavelength measurement : absorbance, transmittance, concentration (Refer to chapter 4-. Wavelength measurement ) At this mode : Measure a sample/samples at preferred / selected wavelength : get the sample s absorbance, transmittance. After applying standard curve, you can calculate sample concentration. - Setup : start wavelength - Wavelength range : 90-00nm 2. Survey scan Mode (Refer to chapter 4-2. Survey scan mode) At this mode : It scans at selected wavelength intervals to measure sample/samples with table and/or graph. Absorbance, transmittance, concentration are measured in each wavelength within selected interval. - Setup : start wavelength, end wavelength, interval - Wavelength range : 90-00nm. PD Sensor PD (Photo Diode) sensor type : PD sensor detects with simple wavelength to measure absorbance of a sample/samples. One detector suggests straight forward detection to reduce defects in test results of several detectors. 2. CCD Sensor CCD (Charge coupled device) sensor type : CCD sensor detects whole spectrum at a time to take results in absorbance of a sample/samples. Suggested to be used to measure sample/samples to proceed Kinetic mode and survey mode at the same time. Time setting Setup starting time to scan. After setting time and scanning intervals, place a sample/ samples into cell-holder, application shall be operated with [Apply]. With setup time, and time intervals, measurement starts with baseline (auto zero) as a reference, and proceeds measuring with whole spectrum. 3-. Select standard curve 3-. Select standard curve 3. Kinetic Mode (Refer to chapter 4-3. Kinetic mode) Select and apply saved standard curve. Standard curve any saved standard curve can be imported (Applied). At this mode : Make time course scanning Find and check results from a sample absorbance s transition according to time flowing and intervals in time. It is suggested to be used to measure a sample that must be tested its reaction in time changes with table and graph. - Setup : start wavelength, end wavelength, interval - Wavelength range : 90-00nm. Touch [Select Standard Curve], Standard curve list will pop up. 4. Hybrid Mode : simple wavelength mode + survey scanning mode (Refer to chapter 4-4. Survey scanning mode and Kinetic mode with CCD sensor : Hybrid Mode) At this mode : Find and check results from a sample absorbance s transition according to time flowing and intervals in time, and also fast scanning to measure a sample on each wavelength in whole spectrum. Hybrid mode is suggested to measure a sample/samples to proceed Kinetic mode and survey mode at the same time

15 2. Select and apply standard curve to measure 3-2. Create standard curve To make new standard curve, measure in setting as below.. Touch [Make Standard Curve] to proceed creating standard curve. 3-. Select standard curve 3. Setting of selected standard curve will be applied automatically. 2. 16 3. Input value of samples concentration. Touch a box of the table or graph that you want to input, a keypad will be appeared. e Check/uncheck on check box then touch [Make Standard Curve] to modify created standard curve. f Confirm a formula and value of R-square in graph at the top. Input mode In case of you already have information regarding concentration and absorbance, input concentration & absorbance directly to make new standard curve 3-2. Create standard curve 4. Insert a sample/samples orderly to proceed measurement. Measure with a multi cell holder a Input the concentration and absorbance of sample. b Touch [Make Standard Curve] to complete making standard curve. c Check/uncheck on check box then touch [Make Standard Curve] to modify standard curve. d Confirm a formula and value of R-square in graph at the top. 5. Touch [Save] to save standard curve Create standard curve a Insert a cell for [Auto Zero] into the [B] cell holder and insert a sample cell into the appointed cell holder. b Touch [Auto Zero] to make auto zero. c After [Auto Zero]. Touch [Measure] to measure. d Touch [Make Standard Curve] to make a new standard curve. e Check/uncheck on check box then touch [Make Standard Curve] to modify standard curve. f Confirm a formula and R-square of graph at the top of graph. Measure with single cell holder a Input the value of sample s concentration and insert a reference cell for [Auto Zero] into cell holder. Then touch [Auto Zero] to make auto zero(reference). b After [Auto Zero], touch [Measure] to proceed. c Repeat measuring to complete sample measurement. d Touch [Make Standard Curve] to make a new standard curve. 30 3

Wavelength measurement (Absorbance:ABS/%Transmittance/Concentration mode) ) Wavelength measurement This mode is simple measurement to measure absorbance, transmittance at selected wavelength.. Input values into start wavelength box and end wavelength, interval, total time and time interval. Measurement method. Insert blank to B (Blank cell holder). Measure reference as a auto zero with [Auto Zero]. 2. After measuring auto zero with blank, ready to set samples. Sure to have samples in cells to each cell holders. [Measure] to applied sample measuring. 3. When there are more samples to proceed survey scanning, process the same as instructed above 2. Repeat [Measure]. 4. Measuring data in table and graph shall be checked screen scroll right and left. 4. Measurement 3. Display and confirm the result of measurement as below. 4-. Wavelength measurement 2 2. 19 2) Wavelength measurement (Standard curve applied) Suggests wavelength measurement to get absorbance, transmittance at selected wavelength and also calculate concentration of sample/samples with standard curve. 3. Touch [Apply] to apply values of selected standard curve to proceed further. 4. Move to measurement display of applied standard curve.. Touch [Select Standard Curve] to apply standard curve. Refer 3-2. Create standard curve to make new standard curve. 4-. Wavelength measurement Iinput name of sample/samples : Touch keypad for inputting name of a sample/samples. Measuring method 4-. Display and confirm the result of measurement as below Survey scan mode ) Survey scan mode Scan and measure selected cell with user's selected interval of wavelength and show all scanning result in chart and table transmittance, absorbance in every each wavelength shall be detected to measure in survey scanning mode.. Input initial wavelength, final wavelength and interval. For an example, display chart and table when measuring starts from 600nm to 700nm with interval 0nm. 4-. 22 4-3. Kinetic mode Measuring method ) Kinetic mode. Insert blank to B(Blank cell holder). Measure reference as a auto zero with [Auto Zero]. Kinetic mode suggests to seek a sample experiment that sees reaction changes of absorbance in time. Table and chart will be shown with measured value.. Input initial wavelength, total time, time interval. For an example, sample will be measured per minitue with 0 seconds interval in 600nm and shown in chart and table. 2. After measuring auto zero with blank, ready to set samples. Sure to have samples in cells to each cell holders. [Measure] to applicate sample measuring. 3. When there are more samples to proceed survey scanning, process the same as instructed above 2. Repeat [Measure]. 4. Measuring data in table and graph shall be checked screen scroll right and left. 3. Display and confirm the result of measurement as below Kinetic Mode Kinetic Mode OPTION Final 2. To show graph and table at a time, [Graph] and [Table] turn into green Survey scanning mode and Kinetic mode with CCD sensor : Hybrid Mode ) Survey scanning mode and Kinetic mode with CCD sensor CCD array sensor is perfect for chemical, biochemical and other applications where fast reactions need to be monitored. Our CCD array sensor mode offers the highest resolution and speed available for Kinetic Absorbance measurements. 2. Input initial wavelength, total time, time interval. For an example, sample will be measured per 5 minutes with 30 seconds interval and 5nm interval from 600nm to 700nm and shown in chart and table Kinetic Mode Name Maximum/Minimum Activity Cursor Instruction The result to measurement shows maximum and minimum value. The result of measurement shows value of Activity. Cursor moves and the position on cursor shows value of the measured result in display. Select PD sensor or CCD array sensor to proceed Hybrid mode. According to the original theory of CCD array, initial wavelength is 220nm and ending wavelength is 780nm Survey scanning mode and Kinetic mode with CCD sensor ABS/%T The value of results are changed to absorbance or transmittance. Setting Graph setup- Peak/Valley setup calues are adjusted by curve s deviation and gradient

To show only table, [Table] turns into green File management ) Open. Touch [Open] to open a saved file, and proceed to file explorer. 30 4-7. File explorer File explorer Change a folder with touch [My Document], [Favorites], [Recent]. 4. Delete file with touch [Delete], cancel the process with touch [Cancel]. 5. [Export] makes files to move to USB flash memory. Touch [Import] to move files from USB flash memory to the equipment. File will be saved automatically in a SD card. Folder creation. Touch [Select] on the left side below on a screen to move a file. 2. Check a file you need to move with touch the file name on the display. 3. With touch [Copy] or [Move] select a folder, and move the file touching [Paste]
