3 CHAPTER 1 Introduction APEL UV-VIS Spectrophotometer PD-3000UV can be used for qualitative analysis and quantitative analysis from the Ultraviolet to Near Infrared wavelength range from nm. The structure of PD-3000UV is simple but stable and accurate, and ideal for use in education, clinical laboratory, biochemistry laboratory, petro chemistry laboratory, environmental protection and various fields of quality control. CHAPTER 2 Safety Instructions 1. Operating temperature should be 5-35, and the humidity should be less than 85%. 2. Put the instrument in an area that is free of strong continuous vibration. 3. Keep the instrument away from the direct sunlight. 4. Keep the direction of air conditioner or electric fan away from the instrument so as to avoid their influence. 5. Keep the instrument away from any electric device that may generate strong magnetic or high frequency electromagnetic field. 6. Confirm the supply voltage and the frequency according to the specifications. And connect earth terminal to the ground. 7. Do not use the instrument in corrosive gases for long time, otherwise it may cause significant shortening of the life of the instrument as well as damage. 8. It is recommended that the instrument is used and stored in clean place. Start and the cautions before the start of the instrument 2.1 Confirm if the environment is suitable for the instrument. 2.2 Before the power cord is connected to the wall outlet, confirm if the position of the voltage change switch suits the supply voltage. And confirm if the supply voltage is normal and the earth terminal is grounded before the use. 2.3 Before the start, make sure if nothing is in the sample compartment and the sample folder is fixed firmly. 2.4 Warm up of the instrument and the initial check-up To ensure accuracy of the measurement, always warm-up the instrument for at least 30 minutes, prior to the operation If it is the first time for the instrument to be used, inspect the stability at %T or Abs range after the warm up. Generally speaking, if the display value fluctuates just like 99.9(0.001),100.0(0.000),100.1(-0.001) at %T(Abs) range, or the least significant digit fluctuates continuously, the condition of the instrument is normal.(it shows the real value because the sensitivity is high.) If the display value fluctuates significantly, consult your supplier When the instrument is not in use for a long time, take the warm up time longer than usual, and observe the stability as new one before the use Prepare some square cuvettes before the test. Clean the inside and outside of the square cuvettes in advance. 7
9 3. Start and check 3.1 Start of the instrument After the power cord is connected, power switch is on and the instrument is started, APEL is displayed on the LCD display. 3.2 System check Few seconds after APEL is displayed on the display, self check is started. The order of the check is FILTER, LAMP CHANGE, SENSOR, D2 LAMP, HALOGEN LAMP, WAVELENGTH CALIBRATION SYSTEM PARAMETER AND DARK CURRENT. Fig. 3.2 Self check System check error When any error is detected, the error alarm sounds, and the error item is displayed at the same time. The check will resume when any key is pressed. Example:The check can not be continued and x is displayed because the dark current is too strong and goes beyond the limit. Press any key to resume the auto-check(see Fig.3.3) Fig. 3.3 System check Dark Current Error (Example) When check error is detected, consult your supplier. 8
10 3.3 System warm up The instrument will remain stable about 20 minutes after the start. And the environment of the halogen lamp will remain stable. After the system check, warm up starts for about 20 minutes. After that dark current test starts the warm up can be stopped by pressing any key. (See Fig. 3.4) Fig. 3.4 System warm up 3.4 Main menu After the warm up, main menu is displayed automatically.(see Fig. 3.5) Fig. 3.5 Main menu Photometry, Quantitative analysis, Qualitative analysis and System setting are displayed as the functions.(see Fig. 3.5) Select one item using up down keys. When is pressed the screen is changed to the selected item. 9

Photometric measurement Absorbance and/or Transmittance at fixed wavelength are measured and displayed or printed out. 4.1 Main menu of the photometry Select Photometry by up down keys in the main menu. When is pressed, the screen is changed to the main screen of Photometry.(See Fig. 4.1) Fig. 4.1 Photometry screen 4.2 Set the measuring mode When SET is pressed in the main screen of Photometry, the screen is changed to measuring mode selection screen.(see Fig.4.2) SET Fig.4.2 Measuring mode selection screen Select measuring modes (Absorbance, Transmittance, Energy) by up down keys. When item)when is pressed the item is selected.(check mark is shown at the right side of each ESC is pressed, the items are selected and the screen is returned to the main screen of the photometry. Note:When Energy is selected and is pressed in the mode selection screen, the screen is changed to Energy screen. 12 4.3 Setting wavelength When GO TO is pressed in the main screen of photometry or measuring screen, the screen is changed to setting screen(see Fig.4.4) Fig. 4.4 Setting Wavelength When the value of the wavelength is input and screen is returned to the main screen of photometry. is pressed, the data is valid and the Note:The valid wavelength range is from 190 to 1100nm, and the data outside of this range is invalid. When the data is invalid, the screen returns to the main screen of photometry after the error alarm sounds. If the input data is wrong, the data can be deleted by pressing CLEAR A/100.0%T(Insert Blank sample to the sample holder) When ZERO is pressed in the main screen of photometry or measuring screen (with blank sample inserted), screen shows 0.000Abs. at Abs mode, or 100.0%T at %T mode Note : Make sure that the blank sample is on the optical path before 0.000Abs/100.0%T is shown. Otherwise the result may be wrong. Please be sure to use quartz square cuvette, if used wavelength is less the 340nm. 4.5 Measurement After 0.000Abs or 100.0%T is shown in the main screen of photometry or measuring screen press and press 4.5) START and move to measuring screen. Make sure the sample is on the optical path,, then the absorbance and the transmittance can be measured.(see Fig. 11
13 START Fig. 4.5 Measured results Note:Only 5 lines of the data are displayed on the screen, and the other data are shown by up and down keys. 4.6 Delete data Press in the measured results screen and select Yes and press, then the measured data is deleted.(memory capacity is 200data max.) If you want to quit deleting data, select No and press E S C CLEAR, or press, then the screen returns to the measuring screen.(see Fig. 4.6) CLEAR Fig. 4.6 Data delete screen 12
14 4.7 Data print out Press PRINT when the measured results are displayed, and select Yes and press, then the printer starts. At the same time when the print is finished, the data are deleted automatically. If you want to quit printing data, select No and press, or E S C, then the screen returns to the measuring screen.(see Fig.4.7) PRINT Fig. 4.7 Data print out screen 13
15 5. Quantitative analysis Concentration of any sample can be measured with the standard curve obtained from standard samples. If the slope and the Y-intercept of the standard straight line are known, the concentration of sample can be measured by coefficient method. 5.1 Main screen of Quantitative analysis Select Quantitative analysis by up down keys in the main menu, and press, then the screen is changed to Quantitative analysis screen.(see Fig. 5.1) Fig. 5.1 Main screen of quantitative analysis 5.2 Standard curve method Standard curve method is a quantitative analysis to know the unknown concentration of any sample with a standard curve obtained from standard samples. 16 5.2.2 Standard curve method screen Select Standard curve method in the Quantitative Analysis screen by up down keys, and press, then enter into the main screen of Standard curve method.(see Fig.5.3) Fig.5.3 Standard curve method Drawing new curve Select Drawing new curve in the main screen of Standard curve method by up down keys, and press, then enter into Drawing new curve procedure. Fill the number of standard samples(see Fig 5.5), press next data screen. Fill the number of data(see Fig. 5.6) and press screen., then enter into Standard sample, and move to the, then move to the next Fig. 5.5 Sample number screen Fig.5.6 Data number screen Note:The range of the number of standard curve is from 1 to 9. Outside of this range is invalid. 15
17 Procedure2 :Set the concentration of the first standard sample. Insert the blank liquid in the optical path, press ZERO first standard sample in the optical path and press sample test screen.(see Fig. 5.7 right) Press screen., set 100%T or 0.000Abs. Fill the concentration of the, then enter into the again and move to the next Fig. 5.7 Setting the concentration of the first standard sample Note:The range of the concentration is from 0 to The outside of this range is invalid. The concentration of the samples to be measured are supposed to be increasing sequentially from the first one to the last one.(the concentration of the first sample must be the lowest.) Procedure 3: Set the second sample, fill the concentration, and press, (the instrument measures the absorbance or transmittance of the standard sample automatically.) Then enter into the next screen.(see Fig. 5.8) Fig. 5.8 Setting the concentration of the second standard sample Note: After setting all the samples and filling all the concentration of the samples according to the designated number of samples and data, the standard curve is drawn automatically. Procedure 4: When all the above procedures are completed, the standard curve is drawn automatically. Press START to enter into the measuring concentration screen to measure the concentration of unknown sample by the standard curve. (See Fig.5.9) 16
18 Fig. 5.9 Drawing standard curve Note: If data error ( operation error or solution error) occurs, the standard curve can not be drawn. And the screen returns to the main screen of standard curve method automatically. Procedure 5:The concentration is measured by the standard curve. After 0.000Abs. or 100%T is set by the blank liquid in the optical path, any sample is set in the optical path. Press Press START PRINT, then the concentration is read and displayed.(see Fig.5.10), then enter into the print selection screen, select Yes, press, then test data and the standard curve are printed out. If No is selected and Fig. 5.11) is pressed, the screen returns to the previous menu.(see Fig Measuring concentration Fig Print selection Setting wavelength Press GO TO in the sample number screen of standard curve method or measuring screen, set the necessary wavelength.(see Fig.5.12) Fig.5.12 Wavelength setting screen Note: Fill the wavelength value in the space under the screen by the numerical keys. Press, then the input is completed and the screen returns to the previous one. Wavelength range of PD-3000UV is from 190 to 1100nm, and outside of the range is invalid. Press CLEAR, and delete the data and try the input again. 17
19 5.2.5 Data delete Press CLEAR Yes and press Fig. 5.13) in the standard curve method, then enter into the data delete screen. Select, all the data on the standard curve method screen are deleted. (See Fig Data delete screen Curve data load Saved standard curve can be loaded from the memory of this instrument.(newly created curve is saved automatically.) When it is necessary, select curve data load by up down keys in the main menu of standard curve method, enter drawing curve selection screen.(see Fig.5.14), then the saved curve data are displayed in the Fig Curve selection screen The newest curve is recorded at the end of the list automatically.(capacity: 200 data max) elect necessary equation, by up down keys in the curve selection screen and press Press START, then the selected curve is displayed.(see Fig.5.15), and the concentration of any sample can be measured by the selected curve. 20 5.2.7 Deleting curves Saved standard curve can be deleted. To delete a certain curve, select Deleting curves by up down keys in the standard curve method screen and press screen., then the screen is changed to Deleting curve Press after a equation is selected, select Yes in the next screen and press, then the equation is deleted. (After the deletion, the screen returns to the previous screen.) Select No in the deleting curve screen and press the screen returns to the previous one.( screen.)(see Fig.5.16) E S C and the deletion is canceled and can be also used to return to the previous Fig Deleting curves 19
21 6. Coefficient method Coefficient method is a simple application of standard curve method. Define the equation and the curve by filling each coefficients of the equation and measure the concentration of any sample by the curve. 6.1 Calculation formula Because the calculation formula of the coefficient method is defined as C=K A+B in the system, parameters K and B are to be filled. 6.2 Coefficient method screen Select Coefficient method by up down keys screen and press in the Quantitative analysis, then move to the coefficient method screen.(see Fig.6.1) Fig. 6.1 Main screen of coefficient method 6.3 Setting parameters screen Set the parameters K and B before the measurement. Press S E T in the coefficient method screen, and the screen is changed to parameter setting screen of coefficient method curve.(see Fig.6.2) Fig.6.2 Parameter setting screen of coefficient method Setting parameter K Select Setting parameter K by up down keys in the coefficient method parameter setting screen and press, then enter into the screen of setting parameter K. 22 6.3.2 Setting parameter B Select Setting parameter B by up down keys in the coefficient method parameter setting screen and press, then enter into the screen of setting parameter B. 23 6.6 Measurement, data output and printout Press START in the coefficient method screen and enter into the measurement screen. 25 8.1 ON/OFF of D2 lamp Select D2 lamp on in the system setting screen and press into D2 lamp ON/OFF selection screen Select ON or OFF in this screen and press ON/OFF is changed.(see Fig.8.3), then the screen enters, then the state of the D2 lamp E S C Fig.8.3 D2 lamp ON/OFF selection screen Note:To keep the life of D2 lamp longer, it is recommended that the D2 lamp is turned off after the start while the test at the UV range is unnecessary for a long time. 8.2 ON/OFF of Halogen lamp Select Halogen lamp on in the system setting screen and press enters into halogen lamp ON/OFF selection screen Select ON or OFF in this screen and press ON/OFF is changed.(see Fig.8.4), then the screen, then the state of the halogen lamp E S C Fig.8.4 Halogen lamp ON/OFF selection screen Note:To keep the life of halogen lamp longer, it is recommended that the halogen lamp is turned off after the start while the test at the VIS range is unnecessary for a long time. 24
26 8.3 Setting Date Select Setting Date in the system screen and press Setting Date screen. (See Fig.8.5), then the screen is changed to E S C Fig.8.5 Setting Date screen Move the cursor by up down keys in the Setting Date screen. The active position goes in the order of Hour, Minute, Second, Year, Month, Date, Day of the week by the key, and it goes in the opposite direction by key. Fill the time and date by numerical keys. As for day of the week, 1 means Monday, 2 means Tuesday,., 7 means Sunday and so on. Press setting screen after the data input, then the date is fixed and the screen returns to the system 8.4 Dark current calibration Dark current should be calibrated before the measurement, because it can affects the results of sample measurement when the wavelength is changed at the wavelength setting or any other reason if the time of dark current is long. Select Dark current calibration screen in the system setting screen and press the calibration starts. If E S C is pressed as instructed on the screen after the calibration, the screen returns to the system setting screen. (See Fig.8.6), then E S C Fig.8.6 Dark current calibration screen 8.5 Wavelength calibration Use this function when the accuracy of the wavelength is investigated. Select wavelength calibration in the system setting screen and press, then the calibration starts. The wavelength is calibrated automatically with the built-in filter (656.1nm). It takes about 2 minutes.(see Fig. 8.7) 25

The range of the wavelength is from 325 to 375nm. 8.7 Initialization Select Initialization in the system setting screen and press changed to initialization screen.(see Fig. 8.9), then the screen is E S C Fig.8.9 Initialization screen 26
28 Select Yes in the initialization selection screen and press, the initialization starts. If No is selected and is pressed, or E S C is pressed directly, the screen returns to the previous one. Note: If Initialization is carried out, all the data in the memory (Test record, parameters, drawn standard curves etc.) are deleted. Please pay close attention when the function is selected. 27
29 Chapter 6 Maintenance Almost no daily maintenance is necessary because PD-3000UV is designed considering durability and reliability. But because there are some parts which have life span such as light source, investigate the performance by calibrating the wavelength or linearity if necessary. And, wipe off dirt frequently, if the dirt is visible. Daily maintenance and cleaning 1) Cleaning of outer case Turn off the power switch and unplug the power cord from the wall outlet. Wipe the surface with a wet soft cloth. If there are stain, use the drop of neutral detergent. 2) Cleaning of cuvette holder Turn off the power switch and unplug the power cord from the wall outlet. Wipe the surface with a wet soft cloth. (If it is necessary to remove the cuvette holder, please consult your supplier.) Replacement of halogen lamp and/or D2 lamp When halogen lamp or D2 lamp is deteriorated, replace them according to the following procedure. 1Turn off the power switch and unplug the power cord from the wall outlet. 2Remove 4 screws of the outer case, disconnect the connectors and open the case. 3Remove the black metal plate which covers the lamp. 4Remove the used lamp from the lamp holder. 5Set new lamp correctly. Caution:Leave the lamp for more than 10 minutes after the power switch is turned off before replacing the lamps, because the lamp can be too hot immediately after the use. D2 lamp should be replaced with the socket, and the halogen lamp should be replaced with the bulb alone. Do not touch the lamp with fingers directly but wrap it with tissue paper or cloth. If the oil from the fingers attaches to the lamp, the measuring accuracy can be deteriorated or the life of the lamp can become shorter. Replacement of fuses The procedure of the replacement is as follows. 1 Turn off the power switch and unplug the power cord from the wall outlet. 2 The fuse holder comes with the power unit together. Therefore insert a flat head screwdriver into the dent and remove the fuse holder. 3 Put a new fuse (2A 5) in the fuse holder and fit it to the original position. Fuse can not be deteriorated in normal use. In case the fuse burns often, there can be any problems, please consult your supplier. Wavelength calibration Calibrate the wavelength according to procedure of above
