3 Formation of micro volume trace,only need 0.5 µl sample Because of liquid tension, the trace sample will be hemispheric which will refract the light and let light can t go through. Make the hemispheric liquid to cylindrical by another force mm height space, sample is cylindrical to enable the light go through.

4 Operation principle of Nano-100 Wavelength will be adjusted by optical filter, then light will go through sample and photodetector, the light will be transfer to electric signal. The absorbance difference between sample and H 2 O, compare with the absorbance of standard sample, so that we get the concentration of sample. 9 Nano-100 clear operation When the measurement is complete, open the upper pedestal and wipe the sample from both the upper and lower pedestals using a soft laboratory wipe. Simple wiping prevents sample carryover in the pedestals

11 Nano-100 software operation instructions Main menu bar Main menu bar includes [File], [Tools], [Debug] and [Help]. [File] includes the following options: Open- Restore the sample data saved in the folder specified by the user, equivalent to My Date key. Save Store result picture of present measurement. Print Print a copy of current data in the default printer. Exit Exit program [Tools] includes the following options: Check correction Test instrument if it is connected properly. Language - Select operation language.

12 Debug option has following functions DebugEnable - enter into Debug mode. LampEnable - open Xenon flash lamp. R_EliminateBlackI - retest black current ContinueBlank - continue the black check automatically ContinueMeasure - continue the measurement automatically SaveBlankData - Save the black date of this measurement Overlay spectra - The user can display 7 different colors (the accumulation sequence is from red, orange, yellow, green, cyan, blue to purple) spectra in the data display area using this feature. After accumulate 7 different colors, they will be cleared. BlankSmooth - Smooth the spectrum data of black check.

14 Nano-100 software operation instructions Functions key bar consist of [Blank] [Measure] [To excel] [To Picture] [ Print]. Blank - Use dissolved sample buffer to make a blank. Before making a sample measurement, a blank must be measured. Measure Initiate the measurement for the samples To Excel The detail absorbance data of present measurement is stored in the Excel table which is specified by user. To Picture To store the current software interface image Print Print a copy of current data in the default printer.

15 The right column of absorbance spectrum includes following information: TypeUsed to select the type of nucleic acid being measured. The user can select DNA-50 for dsdna, RNA-40 for RNA, ssdna-33 for single-stranded DNA. Name-Input the sample name: when sample measurement, the sample name should be input. ID - Display the serial number for the sample being measured. The user can open the software to start recording. A230-Absorbance of the sample at 230 nm with 10 mm path A260-Absorbance of the sample at 260 nm with 10 mm path A280-Absorbance of the sample at 280 nm with 10 mm path
