Integrated data storage and processing approach Global data (zettabytes) Past and projected global data growth (1 ZB = bytes) 1% of global data New approaches should consider high density data storage that is computationally accessible Year Molecules may be one potential solution. For DNA: Ezziane et al. Nanotech., 2006, 17, R27-R39 Gantz and Reinsel (2012). The Digital Universe in Framingham, MA Projected storage densities of Projected processing speeds of bytes/mm 3 ~10 20 reactions/s Data Encode Write Synthesize Store Read Detect Decode Archival Storage Data Access Process Data Distribution Statement A (Approved for Public Release, Distribution Unlimited) 16

What information can molecules currently process? Beyond substantial efforts to mimic Si-based computational approaches (i.e., molecular electronics), there are few examples DNA-based computing on an NP-hard decision problem demonstrates it is conceptually possible to answer computational problems outside of a traditional logic-based approach Start End 1) Assign each node a unique code (address) 2) Incorporate connections (relationships) into code 3) Synthesize and combine all codes 4) Compute: Pairwise associations between nucleotides create DNA strands of various lengths 5) Detect strand lengths, start/end node to determine if a Hamiltonian path exists Adleman, L. Science, 1994, 66, Distribution Statement A (Approved for Public Release, Distribution Unlimited) 18
Molecular diversity offers new ways to represent and process information Millions of molecules exist with unique properties that could be utilized to enable simplified, more capable storage and processing strategies Structure, shape, size, charge, polarity, Potential advantages include: Denser data representation Tunable properties, reactivity and stability Choice of physical representation With a richer feature space for physically representing and querying data, what new operations could molecules enable? Graph analytics Boolean Satisfiability? or (a b c) ( b d a) or Distribution Statement A (Approved for Public Release, Distribution Unlimited) 19
New foundations for information processing Interdisciplinary teams will collectively explore the opportunity space for molecules to store and process information 1) How and what can we encode in molecules? 2) What types of operations can molecules execute? 3) What are the representational abstractions, mathematical or computational primitives, that can describe these operations? 4) What does computation mean in a molecular context? 5) What functions can be decided via molecular means and what equivalence might they have to traditional computing methods? 6) Can we design approaches to compute directly on and with molecular data? Distribution Statement A (Approved for Public Release, Distribution Unlimited) 20
Program overview Proposals will identify mathematical/computational problems to be addressed by the proposed molecular approach Phase 1 (18 mo): Prospects for molecular information storage and processing Encode, read and write: Validate approaches to represent data in molecular form Process: Validate molecular processing approaches against performer-defined problems Phase 2 (18 mo): Strategies to enable direct-access molecular informatics Molecular informatics: Integrate elements to directly process molecular data Anticipated outcomes include: 1) New approaches to represent information and execute computational operations in molecular form 2) Scalable strategies to extract and process information from large molecular data stores 3) Molecular computing concepts that provide capabilities beyond our conventional computational architectures Distribution Statement A (Approved for Public Release, Distribution Unlimited) 21
Specifically excluded are approaches that Mimic current computational architectures (e.g., molecular electronics, molecular logic gates, etc.) or miniaturize chip-based components Provide evolutionary enhancements to past molecular computing efforts Require RNA, enzymes and/or proteins (i.e., biomolecular computing) Utilize nanomaterials (unless as a scaffolding or other support to the molecular information) Cannot be justified as scalable NOTE: DNA-based approaches are within scope, but scalability must be clearly justified Distribution Statement A (Approved for Public Release, Distribution Unlimited) 22
