8 Olis Customization You might have seen exactly what you need on one of the proceeding pages. Or, you might be thinking This one would be great, if only. On these two pages, we show three unique products that we produced for three scientists who knew what they wanted and found us willing to say Yes to producing them. Our ability and willingness to take on custom commissions sets Olis apart from nearly all other spectrophotometry system manufacturers. Rotatable sample chamber for CD Most important to be duplicated: A CD spectrophotometer with a rotatable sample compartment for use in a synchrotron beam. Commissioned by: Guiliano Siligardi, Diamond Light Source. Why needed: A CD spectrophotometer requires linearly polarized light to produce left and right circularly polarized light. The synchrotron produces a highly polarized beam at some fixed angle. The ability to rotate the beam splitting polarizer within the sample compartment allows the optimal matching of the synchrotron s polarization with the beam splitter s requirement of linear light. Short of rotating an entire CD spectrophotometer to achieve the best angle, this design allows Dr. Siligardi to achieve the optimal orientation of the immobile synchrotron beam and thereby obtain the maximum intensity for his CD measurement, while (incidentally) also equalizing the two beams in the Olis DSM 20 CD. 7

9 Olis MultiScan, aka Protein Machine Most anticipated to be copied: A combination instrument bringing together the performance of the Olis DSM 1000 CD, Olis RSM 1000F1, and an Avantes CMOS, fitted with a six position thermostatted cell holder, and novel Data Tree automatic data file storage and retrieval program. Commissioned by: Russ Middaugh, University of Kansas, who christened it the Protein Machine, because it produces a megabyte of data about each protein at every temperature studied. Why needed: Prof Middaugh s group handles hundreds, even thousands of protein samples and hoped to have a single instrument collect all data they wanted for each protein with the minimal variance in the sample s age, temperature, pathlength, and exposure to light from one measurement to the other. Immobilized Enzyme Assay Device Most afield of our normal products: An immobilized enzyme assay device which we designed almost from scratch, after starting with our Automatic Titrator (page 9). Commissioned by: Faical Larachi, University of Laval NEW in 2011 Why needed: Professor Larachi was looking for a modern variant of a device cited in a decadesold publication. We were the one group to say Yes to creating it. We now have a system useful for determining the kinetic parameters of enzymes as they are flushed with substrates, inhibitors, and other reagents. This also exploited our growing interest in utilizing LED as low cost, stable, bright alternatives to traditional light sources. Development of our LED monochromator (page 13) followed soon thereafter. 8
