They provide vertical mounting capabilities for films, gels, sheets of material or multiple filters. By moving the platform across the sample beam you can plot photometric mode versus position in millimeters, with photometric mode being the ordinate and millimeters the abscissa. The Film Holder accessories are fitted with Mylar strips to prevent samples being scratched. All holes in the Cary 4000/5000/6000i version of the accessory are pre-threaded: you need only use thumbscrews. The Film Holder accessory requires the Extended Sample Compartment (part number for Cary 100/300: 0010041800; Cary 400/500/500i: 0010046600; Cary 4000/5000/6000i: 0010079800) and the Sample Transport accessory (part number for Cary 100/300: 0010040600; Cary 400/500/500i: 0010044700). These are available from Agilent. Getting started Unpacking Unpack the accessory and check that you have received all of the components listed in the packing list. For the Cary 100/300 you should have: 1 x Film Holder 4 x V-plates 1 x gel boat mount 2 x plastic clamps with mounting pins 14 x thumbscrews 1 x resting plate 2 x 5 mm magnetic masks 2 x 10 mm magnetic masks 4 x magnetic strips For the Cary 400/500/500i/4000/5000/6000i you should have:

Any damage should be reported immediately. Refer to the accessory by part number. Extended Sample Compartment Sample Transport Accessory 1. Install the Cary 1/3/100/300 Sample Transport accessory according to its instructions. 2. Pull the upper section of the Sample Transport accessory towards you. 3. Hold the Film Holder so that the Mylar strips are on the side to the right. 4. Place it lengthwise on top of the Sample Transport accessory. 5. Line up the holes at each end of the Film Holder with the corresponding holes in the Sample Transport accessory. 6. Fasten with thumbscrews. 7. Slide the Sample Transport accessory back into the sample compartment. 8. Remove the standard base from the Extended Sample Compartment and replace it with the one supplied with the Film Holder. 9. Install the Cary 1/3/100/300 Extended Sample Compartment according to its instructions. Cary 4/5/400/500/500i/4000/5000/6000i You will need: Extended Sample Compartment Sample Transport Accessory 1. Install the Cary 4/5/400/500/500i/4000/5000/6000i Sample Transport accessory according to its instructions. 2. Pull the upper section of the Sample Transport accessory towards you. 3. Hold the Film Holder so that the mylar strips are on the side to the right.
Page 3 of 9 4. Place it lengthwise on top of the Sample Transport accessory, with the lower end of the vertical section towards the back of the Sample Compartment and the higher end towards you. 5. Line up the two smaller holes on the horizontal section of the Film Holder with the two locating pins on the Sample Transport accessory. 6. Push the thumbscrew with the brown standoff through the keyhole and tighten it. 7. Slide the Sample Transport accessory back into the Sample Compartment. 8. Install the Cary 4/5/400/500/500i Extended Sample Compartment or the Cary 4000/5000/6000i Extended Sample Compartment according to its instructions. Accessories V-plates Up to 9 V-plates can be attached underneath each window of the Film Holder. This allows you to, for example, measure a batch of small round filters in turn without having to measure each one individually. Gel boat mounts With the exception of the magnetic masks, all accessories must be attached to the right side of the Film Holder. These hold gel boats in which strips of plate gel can be placed and then scanned. For the Cary 1/3/100/300, one gel boat mount is provided. For the Cary 4/5/400/500/500i/4000/5000/6000i, one gel boat mount is provided for the reference beam and one for the sample beam. Gel boats (part number 6610004200, 8 mm pathlength; 6610004300, 5 mm pathlength) are available from Agilent. Plastic clamps with mounting pins These are used for attaching heavier samples to the Film Holder. The mounting pins can be attached to any hole in the Film Holder, then the clamps attached to them. The clamps may also be used in conjunction with the resting plates.
Page 4 of 9 1. Screw a thumbscrew through the hole in the Film Holder where you want to attach the mounting pin. 2. Screw the mounting pin onto the thumbscrew. 3. Push the plastic clamp onto the mounting pin. The rubber grommets hold the sample in position. Resting plates These are used for supporting heavier samples on the Film Holder. One is provided for the reference beam and one for the sample beam.
Page 5 of 9 1. Attach with thumbscrews beneath the appropriate window of the Film Holder. 2. Rest the bottom of the sample on the resting plate and clamp the top of the sample to the Film Holder. Magnetic masks These allow you to reduce the light beam height if you want to measure only part of a sample. Two widths are provided -- 5 and 10 mm. Place the magnetic masks over the Film Holder's windows, on the left side.
Page 6 of 9 Magnetic strips These are used to attach light, thin samples to the Film Holder. Place a magnetic strip at the top and bottom of the sample to hold it flat against the right side of the Film Holder.
Page 7 of 9 Applications Measuring multiple filters 1. Attach the V-plates underneath each window of the Film Holder, screwing them with thumbscrews onto the hole closest to the base of the Film Holder. The 'V' section should be pointing upwards. The V-plates for the Cary 4000/5000/6000i need one thumbscrew each; those for the Cary 100/300 need two each, placed side-by-side. 2. Rest each filter on the 'V' section of a V-plate. 3. Perform a wavelength scan on each filter in turn. With a filter in each 'V'-holder, the centers of the filters are 20 mm apart. 4. Switch on the computer, then the spectrophotometer and wait for the initialization procedure to be completed. 5. Click the Start button on the Windows taskbar, then Programs, Cary WinUV and Scan. 6. Place the samples on the V-holders. 8. Click the Start button to commence reading. You will also be prompted to select a location in which to store the data files. Each filter will be scanned in turn. Measuring gels You may need to store a separate baseline for each filter position if masks are used. 1. Cut the gel to the correct size to fit in the gel boat. 2. Place the gel in the gel boat. 3. Line up the window on the gel boat mount with the appropriate window of the Film Holder. The lip on the window of the gel boat mount should be at the bottom. For the Cary 1/3/100/300, match up the slot on the top of the gel boat mount with the fifth hole from the front and the second hole from the top of the Film Holder. For the Cary 4/5/400/500/500i, match up the slot on the top of the gel boat mount with the fourth hole from the front of the Film Holder. Because of the positioning of the gel boat mount, you will need to set up the instrument to scan from 15 to 96 mm. 4. Fasten this hole and the two other holes on the gel boat mount with thumbscrews. 5. Place the gel boat on the lip of the window of the gel boat mount. The rubber grommets at each end of the window hold the boat in place. 6. Click the Start button on the Windows taskbar, then Programs, Cary WinUV and Scan. 7.
