1 Name: Date: Microscope Labs #1 and #2 e Lab and Hair Lab Day 1 - e Lab Purpose: To study the image formed by the microscope Procedure: You may begin once you go to the lab area! A. Preparation of the slide (This is done on your table!) a. Place a drop of water in the center of the slide b. Cut out a lowercase e from the body of a newspaper article c. Place the e on the drop of water on the slide d. Cover the e with a cover slip B. Adjustment of the microscope Adjust your microscope following the Focusing the Microscope directions that you have already received. Be sure you are starting on low power! C. Examination of the slide a. Place the slide on the stage of the microscope so that the e is right side up when you look at it, with your eyes, from behind the arm of the microscope. b. Center the slide so the e is directly over the hole in the stage. c. Look through the ocular and slowly turn the coarse adjustment knob until the image is clear. Sharpen the image by using the fine adjustment knob. d. While looking through the ocular, center the image by moving the slide. e. Carefully draw the centered image exactly as you see it. f. While watching the objectives, slowly turn the revolving nosepiece until the high power objective clicks into place. g. Use the fine adjustment knob ONLY to sharpen the image. h. Carefully draw the image exactly as you see it. i. Go back to low power by turning the nosepiece until the low power objective clicks into place. j. While looking through the ocular, push the slide away from yourself. Observe the movement. Record the movement. k. Push the slide to the right. Observe the movement. Record the movement. Observations: Use the following list to ensure that you have d all necessary observations. Check off each one as you it. A. Carefully draw the image exactly as you see it under low power (See above C. e.) B. Carefully draw the image exactly as you see it under high power (See above C. h.) C. Both drawings should be done using pencil or colored pencils, drawn inside of a traced circle and labeled with the total magnification below the circle. D. Record the movement of the slide when you pushed it in step C. j. & C. k.

2 Day 2 - Hair Lab Purpose: To observe depth of field using overlapping hairs under the microscope Procedure: You may begin once you go to the lab area! A. Preparation of the slide a. Place a drop of water in the center of the slide b. Cut a small piece of yellow thread (approximately 2 cm.) and place it on the drop of water on the slide. c. Cut a small piece of your hair (approximately 2 cm.) and place it on top of the yellow thread to form an. d. Cover the of the hairs with a cover slip. B. Adjustment of the microscope Adjust your microscope following the Focusing the Microscope directions that you have already received. Be sure to start on low power! C. Examination of the slide a. Place the slide on the stage of the microscope. b. Center the slide so the is directly over the hole in the stage. c. Lower the body tube. d. Look through the ocular and slowly turn the coarse adjustment knob until the image is clear. e. While looking through the ocular, center the image by moving the slide. You may need to do this several times to correctly answer the following questions. i. Does the thread or hair come into focus first? ii. Was this placed on top or bottom? iii. When your hair is in sharp focus, can you see the yellow thread? iv. When your hair is in sharp focus, is the thread also in sharp focus? h. Carefully draw the image exactly as you see it. i. While watching the nosepiece, slowly turn it until the high power objective clicks into place. j. Use the fine adjustment knob ONLY to sharpen the image. Answer the following questions. i. Does the hair or thread come into focus first? ii. Was this placed on the top or bottom? iii. When the thread is in sharp focus, can you see your hair? iv. When the thread is in sharp focus, is your hair also in sharp focus? k. Carefully draw the image exactly as you see it.

Both drawings should be done using pencil or colored pencils, drawn inside of a traced circle and labeled with the total magnification below the circle. D. Record the movement of the slide when you pushed it in step C. l. E. Be sure to write down the answers to the questions in C. g. and C.j. Conclusions: All conclusions should be written with a pen or typed and should be paragraph form. Be sure to include the answers to all of the questions! A. Write a brief (3-4 sentences) overview of what you did in class to both lab activities. B. Day 1 a. What did we see? i. Compare the e as your eye sees it to the image seen under low power. Compare the image of the e seen under low power with the image of the e seen under high power. Describe the paper itself (not the ink). ii. Recall the movement of the slide from lab and describe how the image moved in your field of view when you moved the slide. When the slide is pushed to the right, what way does the e appear to move? When you move the slide away from yourself, what way does the e appear to move? Based on your observations, what can you conclude? C. Day 2 a. Low power i. Is the thread or hair in sharp focus first? What can you conclude about how the microscope focuses? Are both in sharp focus at the same time? b. High Power i. Is the thread or hair in sharp focus first? Are both in sharp focus at the same time? What can you conclude? Which section of the low power field is also seen under high power. D. Does high or low power require more light? Look carefully at your microscope. Why does this power require more light? Were you able to see depth under the microscope? What must you do to assure that you see all levels of the object you are observing? Describe, in detail, how to focus under high power. Start with The image is perfectly focused on low power E.
