2 Olympus Digital Microscope Camera (DP70) checklist Camera Information - Still images can be obtained using the Olympus DP70 camera Startup - Remove dust cover from Olympus BX51 compound microscope to which the Olympus DP70 camera is attached - If you are going to use the Olympus dissecting microscope, do the following: - Remove dust cover from the dissecting microscope - Remove dust cap - The Olympus DP70 camera is attached to the BX51 Olympus microscope with a U- PMTVC adapter. Use the Allan wrench that comes with the microscope to loosen the lower screw of the adapter. CARFULLY remove the Olympus DP70 camera with adapter from the Olympus BX51microscope and place it onto the dissecting microscope - Use Allan wrench to tighten lower screw of the adapter - If you are going to use the Olympus CH-2 compound microscope, do the following: - Remove dust cover from the CH-2 compound microscope - Remove dust cap - The Olympus DP70 camera is attached to the BX51 Olympus microscope with a U- PMTVC adapter. Use the Allan wrench that comes with the microscope to loosen the lower screw of the adapter. CARFULLY remove the Olympus DP70 camera with adapter from the Olympus BX51microscope and place it onto the CH-2 microscope - Use Allan wrench to tighten lower screw of the adapter - Turn on computer monitor - Turn on computer - A login screen (Novell Client for Window) appears - Select Workstation only if you do not want to be connected to the server. A User Name appears. Type in the password and select OK - Uncheck Workstation only if you want to be able to connect to the server. Type in your User Name and Novell password. Select OK. A Windows Workstation window opens and a User Name appears. Type in password and click OK 1
3 Retrieve an Image Microscope Setup - If using a compound microscope: - Turn microscope ON - Find specimen on slide using the microscope and change objective if needed - Set Köhler illumination to increase quality of obtained image. NOTE: Use Operating Checklist for Olympus BX51 for detailed instructions on how to setup the Olympus BX51 microscope - Pull out level with black knob on right side of oculars (sent light to the camera) - If using a dissecting microscope: - Turn microscope ON - Find specimen using the dissecting microscope - Set level on the right side of microscope to PHOTO and IN to sent light to the camera - Click on DPController icon. This opens an Olympus window to control the camera 2
4 Capture - Select the CAPTURE tab - Click on the eyeball with the bleu arrowhead (lower left) to get a live image on the computer screen. To focus image use focus knob on the microscope - Box (spot) selects area used for exposure calculations. Change the size of the exposure box (30%, 1%, 0.1%) in the Spot group - Make adjustments for size of image in the Still-image group. When image size 2040x1536 or 4080x3072 is selected in the Still-image group, pixel shifting (method for obtaining images with higher resolution) is performed. The time to capture an image will increase 9 times compared to the current exposure time (displayed in Still-image group) - To adjust exposure settings, make changes in the Exposure time group, Sensitivity group and Exposure group - To change exposure (slider in the Exposure group), select Manual in the Exposure Mode group. To increase (recommended for dark specimen) and decrease (recommended for bright specimen) the exposure in steps (bracket), select the correct adjustment in the Exposure time group - For bright-field, DIC, phase contrast, select Auto in Exposure Mode group - For fluorescence, select SFL-Auto in Exposure Mode group - When increasing sensitivity (higher ISO values) the exposure time is shortened. Note that this may increase the noise in your captured image - If a scale bar is displayed in your image, make sure the correct objective is selected in the Objective group - To decrease noise in an image captured from a noisy observation image, select the Average mode and specify a count in the Accumulation group - to provide contrast between the background of an observation image and the observation target region during fluorescence observation, select the Integral mode and specify a count in the Accumulation group. Adjust size of exposure box to the size of the target region to increase contrast in the captured image even more - To take a picture click on the camera icon. Your image will appear in DPManager - Save Images to your folder in the tempimages folder on the desktop or to another medium. Make a folder with your own name inside the tempimages folder if you don t have one. Make sure you back up your images regularly and delete them from your folder. THIS COMPUTER IS NOT A MEDIUM TO STORE IMAGES Preview - If an observation image is dark, the sensitivity can be increased to brighten the image. Use 2x2 or 4x4 in the Binning group to increase sensitivity 4 times and 16 times respectively. Note that the resolution of the observed image will decrease but the captured image will not be effected - If an observation image is noisy, the noise can be reduced by applying frame averaging selecting 2 frame in the Real-time frame average group. Note that this feature only reduces noise in the observation image and is not applied to captured images 3
5 Color Balance - Select COLOR BALANCE tab to correct for white balance (mainly for bright-field) and black balance (mainly for fluorescence) WHITE BALANCE - Select On and One Touch to adjust the color so that the color of the specified region (dropper) in the observation image is white - Select On and One Push to adjust the colors in the observation image so that the entire image looks white - Select On and adjust color balance levels for the color channels (Red, Green, Blue) using sliders BLACK BALANCE - Select On and One Touch to adjust the brightness of the observation image so the specified region (dropper) in the observation image is black - Select On and One Push to adjust the brightness of the observation image so that the entire image looks black Select On to adjust the brightness of the entire observation image using a slider Level Adjust - Select LEVEL ADJUST tab to image contrast by changing the intensity levels of the individual color channels (Red, Blue, Green). After adjustment per channel, combined intensity levels of the color channels can be adjusted User Setting - Use the USER SETTINGS tab to save software settings in order to use them again later - Save your User settings file in your folder inside the tempimages folder on the desktop - To load previous saved User settings, select load in de User settings group. Find and select your user settings and select open to apply user settings. See attachment I for information that is saved Scale - Select SCALE tab if you want to get a scale bar in your image - Check Show Scale in Display to add scale bar to your image - Select Free Size in Display to rotate or to adjust the size of the scale bar - Select Include in Image in Display for scale bar to show up in captured image - Select desired bar shape and add the size by selecting number style - To get the correct scale bar select the objective you use (circle w/ objectives) and make sure the adapter lens is set to 0.75x (0.3 mount X 2.5 photo eyepiece) - To change color or other features of the scale bar, place cursor on scale bar and right click on the mouse button 4
6 Time Lapse - Select TIMELAPSE tab if you want to record changes over time as a movie - Check if the Capture in timelapse mode in selected - In the figure below the relationship between Time to start capture, Interval of capture and Capturing time is shown: - Set the hours, minutes and seconds of Time to start capture using the up and down buttons - Set the hours, minutes and seconds of Interval of capture. This is the time between each image captured - Under Number of capture, set the total number of frames you want to collect - Check the Capture as AVI checkbox - Select the CAPTURE tab - Open the Size dropdown list and select the image size to be captured. NOTE: The maximum image size of the recorded movie is 1360 x Click on the capture button. The recorded movie is sent to the DP Manager 5
7 Top Row Functions (from left to right) - NORMAL - full field of view - ZOOM center of images is zoomed twice - FULL The resolution of the CCD is the whole screen displayed - FULL SCREEN Image is enlarged and displayed full screen on the desktop. Right click mouse button to get back to previous window - OVERLAY FLICKERING RESTRAINT The flickering of overlay graphics is restrained - BAR GRAPH DISPLAY Display focus indicator as bar graph - PROFILE Display focus as profile. An intensity profile (RGB) along the gray line in image is shown - NUMBER DISPLAY Display focus indicator numerically ( Current and Max ) - FOCUS RESET Reset focus indicator value to adjust Max value to Current value (only if Number Display is selected) - ONE TOUCH WHITE BALANCE Color is adjusted so that the color of the specified region (dropper) in the observed image is white - ONE TOUCH BLACK BALANCE Brightness of the observation image is adjusted so that the specified region (dropper) in the observation image is black - ONE PUSH WHITH BALANCE Colors in the observation image are adjusted so that the entire image looks white - ONE PUSH BLACK BALANCE - Brightness of the observation image is adjusted so that the entire image looks black - ROTATE 180 Image is rotated FLIP HORIZONTALLY Flip image horizontally - FLIP VERTICALLY Flip image vertically - GRAYSCALE The color display is switched to gray scale - CLIP REGION Clip region is specified. Adjust size if needed - TEXT INPUT Add text to your image. Go to image and select an area (click and drag mouse). An Edit Text window opens to write your text - APPEND TO STILL IMAGE The text in the image preview will be included in the captured image (only if Text Input is selected) - 16 BITS CHANNEL The bits of color per channel of the image is changed from 8 to 16 - MINIMUM SHARPNESS The image is sharpened lightly - NORMAL SHARPNESS The image is sharpened moderately - MAXIMUM SHARPNESS The image is sharpened heavily - HELP Launches DP Controller Help window in case you have questions about the different functions or buttons 6
8 Image Composition Overlay - Open all images you want to overlay in DP Manager. Go to tempimages folder and drag desired images into DP Manager window - Select the Image Composition icon to display the Image Composition window È - In the mode group, select Normal - In the image group, select a composite candidate image in the dropdown menu. The checkbox in the left of a composite candidate image list is checked. To clear a selected image, click on the check box in the left of the drop-down list - To fine adjust the position of the composed image, click on the up and down buttons in the XY Position group - Adjust the intensity of the brightness of each image by using the up and down buttons in the Colors/Intensities group - To increase zoom, use the slider in the zoom group or enter the desired zoom ratio directly in the box below the slider. Selecting Fit to window button displays the image filling the preview area. When selecting the 100% button, each of the image pixels will be represented by a monitor pixel - If the previewed composite image at the top left of the window is acceptable, select Apply. The overlay image will appear in the DP Manager Shut Down - Did you save your image(s)? - Close Olympus window and DP Manager window - Go to START, select SHUTDOWN and select OK in the shutdown window - Wait for the computer to shut down - Turn of the monitor - Remove the sample and turn of the microscope (default setting Æ bright-field) - If you attached the camera to the Olympus CH-2 microscope or the dissecting microscope, make sure to remove the camera w/adapter and attach it to the Olympus BX51 microscope - Leave the room CLEAN and TIDY.AS ALWAYS!!!!
