2 General Infrmatin COURSE OBJECTIVE The primary bjective f this curse is fr students t learn labratry methds fr characterizing bilgical macrmlecules using biphysical appraches. EQUIPMENT AND SUPPLIES REPLACEMENT In the event that glassware r ther equipment and supplies are damaged r brken by students, the student must pay fr the replacement f thse items with yur Buzz card. 3 ATTENDANCE Students are expected t attend all lectures and reprt t class n time. Lack f attendance and/r tardiness t lab, and tardy assignments will be excused nly with prmpt written dcumentatin t the satisfactin f the instructr. Due t limitatins in space and supprt persnnel, pprtunities t make up missed labratry experiments will nt be pssible. Grading The verall grading scheme fr this curse is based n the fllwing scheme. Labratry Reprts 30% (lwest reprt grade drpped frm average) Labratry Perfrmance 30% Hmewrk 10% Examinatins 10% Ntebk Maintenance 10% Experiment Presentatin 10% Final grades will be cmputed based n the grading scheme abve. Final letter grades will be issued accrding t the fllwing delineatin: A % B 80 89% C 70 79% D 60 69% F <60% 1. LABORATORY REPORTS The purpse f the labratry reprt is t cmmunicate experimental wrk in writing. The educatinal gal is t help students learn and practice expressing their ideas and describing their wrk in a prfessinal manner. With this in mind, the requirements fr the structure f the labratry reprt are similar t thse fr peer-reviewed scientific literature: Labratry reprts must be submitted in hard cpy by 1 pm n the due date. N late lab reprts will be accepted. Acceptable labratry reprts will be n lnger than 10 duble-spaced pages with writing n ne side f the paper (title page nt included). NOTE: Duble-spacing allws graders t make cmments near the relevant text. The prject prpsal can be as lng as 15 duble-spaced pages with wrd-prcessing n nly ne side f the paper. Reprts shuld be written in grammatically crrect English, and prepared using a clear fnt f size 12-pint, with 1-inch margins n all sides. Each page shuld be numbered at the bttm. Labratry reprts shuld NOT be frmatted with dual clumn text as seen in published jurnal articles. 3

4 Figures shuld have figure legends describing the figure in sufficient detail underneath the figure. Tables shuld have headings at the tp f the table. Fnt sizes f text fr figure legends r tables may be 8-10 pint in size and figure legends r table-heading text may be single-spaced. Labratry Reprt Cmpnents Cver Page (Required) Each lab reprt shuld have a cver page that cntains the title f the experiment, the name f the student, the name f any labratry partner, the dates f the experimental wrk and the date submitted. Als include a signature stating cmpliance with the Gergia Tech Hnr Cde. Intrductin (15 pints) Present backgrund fr the experimental wrk described. State relevant cncepts and hyptheses and the bjectives f the experiment. Refer t jurnal articles (nt web sites) where yu have read supprtive backgrund infrmatin. An intrductin is nly required fr full-length lab reprts. Experimental Prcedures (15 pints) Summarize the specialized reagents and their surces and equipment used in the experiment. Generally describe methds used especially where deviatins t the prtcl were made. Describe the experimental prcedures in the level f detail cmmnly fund in published research articles and, nly when necessary, add additinal details. The experimental prcedures shuld be described in bth shrt and full-length lab reprts, with mre details in the full-length reprts than in shrt reprts. Results (30 pints) Describe the data generated frm the experiment in wrds. Then, present figures (including graphs) r tables f yur data fr emphasis and clarity. Each figure shuld have a figure legend underneath the figure - a statement describing the figure itself. Each table shuld have a table heading abve the table. All figures shuld be clearly labeled. Results sectins with insufficient text describing the results and/r key illustratins f data will have severe pint deductins. Results shuld be described in bth shrt and full-length labratry reprts. Discussin (30 pints) State the verall cnclusins frm yur experiment here. In cases where the wrk was hypthesis-based, the discussin shuld address the hypthesis directly. Discuss the significance, implicatins, cmparisns f results t ther wrk, etc. State and critically evaluate any assumptins that were made. Estimate the accuracy f yur results. Discuss any bservatins that yu fund unusual r unexpected, and why they may have ccurred. Nte and discuss incnsistencies in yur data that make drawing firm cnclusins difficult. Discuss imprvements that culd be made t ptentially imprve yur results. A discussin sectin is required fr full-length labratry reprts, and a brief cnclusin replaces the discussin sectin fr shrt labratry reprts. References (10 pints) List the references made thrughut the text f the research article in the rder in which yu refer t them in the text. References are required, nt ptinal! Reputable resurces shuld be used as references. Web sites are nt reputable resurces fr a prfessinal labratry reprt. Avid referencing yur Bichemistry textbk since it cntains general infrmatin. 4

5 2. LABORATORY PERFORMANCE/CONDUCT Students are expected t have read the labratry prtcl fr the day prir t reprting t class. Nte: Be sure t review all links assciated with a labratry prtcl. Students are expected t fllw written prcedures fr cnducting assigned experiments. Due t limitatins in equipment, students will be asked t wrk with the TA/instructr in cycling thrugh the labratry wrk. Students are expected t wrk independently (r when necessary, with labratry partners). Students are respnsible fr the upkeep f their assigned wrkstatins. Be sure t restck all pipette tip bxes at the end f each class perid. Be sure t clean all glassware befre the end f the class perid. Keep the balance and instrumentatin areas clean and free frm clutter. Be sure t reprt any malfunctins in equipment t the TA r instructr. All materials stred in the refrigeratr, freezers, r at rm temperature must be capped and clearly labeled with yur name and its cntents. Fllw all safety regulatins and encurage thers arund yu t wrk safely as well. D nt eat, drink, r chew gum in the labratry. D nt bring fd, including bttled water fr drinking int the labratry. D nt discard fd trash in the labratry. D nt bring fd int the labratry. Labratry Hazards Sme f the chemicals used in this labratry are harmful if inhaled r ingested. New Plicy in Fall 2011: Students must wear a labratry cat at all time while wrking in the Bichemistry Teaching labratry. The cat may be made f a 65%/35% plyester/cttn blend, 100% cttn, r any higher-grade safety fabric. Typical lab cats being sld n campus are full-length. Aprns will nt suffice. Other requirements are described in detail in the Persnal Prtective Equipment and Labratry Attire Plicy (See T-Square Resurces fr PDF). Always wear safety glasses in the Bichemistry Labratry! Reading glasses n lnger suffice as suitable safety prtectin fr the eyes. Wear suitable clthing in the Bichemistry Labratry. Sandals and shrts are nt permitted in the lab. Tes must be cvered. Wear dispsable latex r nitrile glves when wrking with dangerus chemicals. D nt allw labratry chemicals t enter yur muth r small cuts r scratches n yur hands. Dispsable glves are available fr daily use t avid this prblem and t prhibit cntaminatin f labratry experiments. 5

7 3. HOMEWORK There will be 5 hmewrk assignments ver the curse f the term. Hmewrk assignments will be psted n T-Square and will typically be due in hard-cpy frmat at the beginning f the lecture perid. Hmewrk due dates are prvided in the schedule. 4. EXAMINATIONS There will be tw midterm examinatins given this semester. Attendance is mandatry n thse dates. The examinatins will be clsed bk (unless therwise specified). Students may use a standard calculatr, if necessary. Calculatrs may nt be shared. Examinatins will cver material frm lecture and the labratry sessins cvered prir t each examinatin. Exam #2 is essentially cumulative! 5. LABORATORY NOTEBOOK MAINTENANCE All experimental data, except instrument utput, shuld be recrded in indelible ink in a bund labratry ntebk with pre-printed sequential page numbers. Students shuld sign the ntebk n the last page f that day s experiment (minimally). It is best t sign each ntebk page, thugh. D nt leave blank pages in a labratry ntebk. A lab ntebk shuld include: Experiment title Date n which wrk was cnducted Gal what is the purpse f the experiment Materials used, identificatin f samples, surces f key reagents, manufacturers f specialized equipment Experimental prcedures perfrmed in sufficient detail t reprduce results Experimental results and bservatins Relevant data analysis (ptinal and highly useful, but nt rigrusly required) Preferably a cnclusin statement Fr example, Experiment was successful r FAILED: D nt repeat as written Signature with date Recrd data and bservatins as yu btain r make them. D nt write n scraps f paper with the intentin f transferring infrmatin t the lab ntebk later. D nt wrry if yur ntebk is a little messy. The recrding and rganizatin f a permanent recrd f labratry bservatins is as imprtant a technique t master as any f the experimental methds yu learn. The research ntebk is a day-by-day recrd f the prgress f experimental wrk. It shuld reflect the integrity and hnesty f the experimenter as well as the clarity f his r her thught. 6. PRESENTATION Student will give a shrt presentatin n ne f the last experiments cnducted in the curse. Mre details abut this assignment will be given later in the term. Presentatins and their evaluatin will be similar t thse given in CHEM 4581 fr the GMO prject. 7
